Upregulation of LGALS1 is associated with oral cancer metastasis

Abstract: Background:Oral cancer metastasis is a devastating process that contributes to poor prognosis and high mortality, yet its detailed underlying mechanisms remain unclear. Here, we aimed to evaluate metastasis-specific markers in oral cancer and to provide comprehensive recognition concerning functional roles of the specific target in oral cancer metastasis. Methods:Lectin, galactoside-binding, soluble, 1 (LGALS1) was identified by secretomic analysis. LGALS1 expression of patient samples with oral cancer on the t… Show more

“…TSPAN15 is believed to enhance tumor stemness, and it has the ability to promote tumor growth and recurrence [30]. TSPAN12 can promote tumor cell proliferation and colony formation, and high expression of CD9 is considered to be related to lymph node metastasis [31,32]. In contrast, another portion of the four-transmembrane protein family showed tumor suppression.…”

“…After the membranes were blocked with 5% (w/v) skimmed milk or BSA (bovine serum albumin) in Tris-buffered saline with Tween-20 [TBST; 50 mM Tris, 150 mM NaCl, and 0.1% Tween-20 (v/v); pH 8.0] for 1 h, the primary antibodies (Genetex Inc., Hsinchu, Taiwan, Table 1) were diluted 1:2000 and added onto the membranes, which were incubated overnight to 48 h at 4 • C. Following this, the membranes were washed in TBST for 6 times (10 min/wash) and then incubated in TBST solution containing appropriate horseradish peroxidase-coupled secondary antibodies (Jackson ImmunoResearch Laboratories, Inc., West Grove, PA, USA) in dilution of 1:10,000 for 1 h with gentle agitation. Then, the membranes were washed again in TBST 6 times (10 min/time), and the probed proteins were visualized using an enhanced chemiluminescence (ECL) method (Visual Protein Biotech Corp., Taipei, Taiwan) [42]. The data of densitometric quantification based on ratio to housekeeping protein (LDH) were evaluated by Image J software.…”
