Upregulation of Arc mRNA expression in the prefrontal cortex following cue‐induced reinstatement of extinguished cocaine‐seeking behavior

Abstract: Cocaine-associated cues acquire incentive motivational effects that manifest as cue-elicited craving in humans and cocaine-seeking behavior in rats. Here we examine the hypothesis that neuronal processes associated with incentive motivational effects of cocaine cues involve increased expression of the plasticity-associated gene, Arc. Rats trained to self-administer cocaine subsequently underwent extinction training, during which cocaine-seeking behavior (i.e., responses without cocaine reinforcement) progressi… Show more

“…These findings expand upon our previous study (Lasseter et al, 2011), in which BM-induced functional disconnection of the OFC and BLA inhibited drug context-induced reinstatement of cocaine-seeking behavior without altering food-reinforced instrumental responding or general motor activity. These findings also add to a larger literature that indicates that the OFC and BLA are critical for explicit cue-and drug context-induced cocaine-seeking behaviors (Atkins et al, 2008;Crombag et al, 2008;Fuchs et al, 2005Fuchs et al, , 2004Fuchs et al, , 2008Grimm and See, 2000;Hamlin et al, 2008;Kantak et al, 2002;Lasseter et al, 2009Lasseter et al, , 2011McLaughlin and See, 2003;Neisewander et al, 2000;See et al, 2001;Zavala et al, 2008).…”

“…The lack of changes in Fos expression in these regions does not exclude a role of AMPA receptors since AMPA signal transduction in these regions may involve molecular pathways that induce other IEGs, such as Arc or Zif268 that are also activated by drug-associated stimuli (Hearing et al, 2008;Koya et al, 2006;Schiltz et al, 2005;Thomas et al, 2003;Zavala et al, 2008). Collectively, the present findings suggest a region-specific involvement of AMPA receptors in cocaine-conditioned c-fos induction.…”

“…Fos in PL neurons is also increased on the last day of cocaine self-administration (Zavala et al, 2007), the first day of extinction (when seeking levels were high) (Nic Dhonnchadha et al, 2012), and during cocaine-associated context discriminative or discrete cues that produced reinstatement (Ciccocioppo et al, 2001; Zhou et al, 2014). PL ARC mRNA is also elevated in cue-induced reinstatement (Zavala et al, 2008). After a period of cocaine abstinence, which has the impact of incubating, or upregulating craving for cocaine (Grimm et al, 2001), c-fos , zif268 , and ARC mRNA are all elevated in PL (Hearing et al, 2008).…”
