Updated Epstein–Barr virus (EBV) DNA sequence and analysis of a promoter for the BART (CST, BARF0) RNAs of EBV

Abstract: Two sequences required for activity of the Epstein-Barr virus BART RNA promoter in transfection assays have been identified by site-directed mutagenesis. One contains a consensus AP-1 site; the other has some similarity to Ets and Stat consensus binding sites. Candidate sequences were suggested by mapping a region of unmethylated DNA in EBV around the BART promoter followed by in vivo footprinting the promoter in the C666-1 nasopharyngeal carcinoma cell line, which expresses BART RNAs. The data are presented i… Show more

“…According to the newly revised EBV sequence (12) (GenBank accession number AJ507799), the predicted coding sequence of BKRF3 contains 765 nt and encodes a protein of 255 amino acids. The predicted amino acid sequence of BKRF3 was aligned with different eukaryotic and prokaryotic Ung sequences (Fig.…”

“…Epigenetic status of the EBV BART promoter region has been analyzed by several investigators previously (Smith et al, 2000;de Jesus et al, 2003;Chen et al, 2005;Al-Mozaini et al, 2009). One group reported that BART promoter activity was found in about 1 kb region encompassing just upstream of the transcription start site to a distance downstream of exon I of the BART gene (de Jesus et al, 2003;Al-Mozaini et al, 2009).…”

“…CpG sites are indicated with vertical ticks, Taq I restriction site is indicated with the vertical triangle, and PCR primers for COBRA analysis are indicated with arrowheads. BART promoter regions analyzed previously (de Jesus et al, 2003;Chen et al, 2005) Table S1, are plotted in a bar graph for easy comparison. be produced as Cp/Wp are silenced due to DNA methylation (Li and Minarovits, 2003;Tao and Robertson, 2003).…”
