This study aimed to compare the efficacy and safety of traditional Chinese medicines (TCMs) combined with paclitaxel-based chemotherapy and paclitaxel-based chemotherapy alone for gastric cancer treatment. Literature searches (up to September 25, 2019) were performed using the Cochrane Library, EMBASE, PubMed, Chinese Science and Technology Journals (CQVIP), Wanfang, and China Academic Journals (CNKI) databases. Data from 14 randomized controlled trials (RCTs), with 1,109 participants, were included. The results indicated that, compared with paclitaxel-based chemotherapy alone, the combination of TCMs and paclitaxel-based chemotherapy significantly improved the tumor response rate (TRR; RR: 1.39; 95% CI: 1.24-1.57; p < 0.001, I 2 = 12%), increased the quality of life based on the Karnofsky Performance Scale score (RR: 1.53; 95% CI: 1.19-1.96; p < 0.001, I 2 = 0%), and reduced the side effects, such as neutropenia (RR: 0.68; 95% CI: 0.55-0.84; p < 0.001, I 2 = 44%), leukopenia (RR: 0.69; 95% CI: 0.54-0.90; p < 0.01, I 2 = 40%), thrombocytopenia (RR: 0.66; 95% CI: 0.46-0.96; p < 0.05, I 2 = 32%), and nausea and vomiting (RR: 0.50; 95% CI: 0.32-0.80; p < 0.01, I 2 = 85%). Hepatic dysfunction (RR: 0.63; 95% CI: 0.33-1.20; p = 0.16, I 2 = 0%), neurotoxicity (RR: 0.64; 95% CI: 0.26-1.55; p = 0.32, I 2 = 0%), and anemia (RR: 0.65; 95% CI: 0.40-1.04; p = 0.07, I 2 = 0%) were similar between the two groups. Evidence from the meta-analysis suggested that compared with paclitaxelbased chemotherapy alone, the combination of TCMs and paclitaxel-based chemotherapy may increase the TRR, improve quality of life, and reduce multiple chemotherapy-related side effects in gastric cancer patients. Additional rigorously designed large RCTs are required to confirm the efficacy and safety of this treatment.

Background X antigen family member 1B (XAGE-1b), a member of XAGE subfamily and GAGE family, is upregulated in some malignant tumors and has been associated with the proliferation, invasion and metastasis of tumors. However, the biological roles of XAGE-1b in gastric cancer (GC) still remain unclear. Methods We detected the expression of XAGE-1b in 60 paired fresh tissues of GC patients by real-time RT-PCR. Kaplan-Meier survival curve was explored to analyze 5-year survival time of GC patients. Function experiments were performed to estimate the role of XAGE-1b on the proliferation, invasion and metastasis of GC cells. Informatic analysis was applied to investigate the potential mechanisms. Results XAGE-1b was obviously upregulated in GC tissues. XAGE-1b was correlated significantly with poor prognosis of GC patients. XAGE-1b markedly promoted the proliferation and invasion in GC cell lines in vitro. Knockdown of XAGE-1b promoted the pulmonary metastatic ability in nude mice. Moreover, XAGE-1b was positively or negatively correlated with the expression of CLDN6 and CHGA, which regulated the progression of GC. Conclusions XAGE-1b could act as an oncogene in GC, which provides a potential biological marker or treatment target for GC.

Discovery of novel isoliquiritigenin analogue ISL-17 as a potential anti-gastric cancer agent

Isoliquiritigenin (ISL), a natural product isolated from licorice root, exhibits anti-gastric cancer effects. However, applications of ISL are still limited in clinical practice due to its poor bioavailability. To discovery of more effective anti-gastric cancer agents based on ISL, aldol condensation reaction was applied to synthesize the ISL analogues. MTS assay was used to evaluate the inhibitory activities of ISL analogues against SGC-7901, BGC-823 and GES-1 cells in vitro. Cell cycle distribution, apoptosis and reactive oxygen species (ROS) generation were detected by flow cytometry. Western blot assay was used to analyze the expression levels of related proteins. The drug-likeness and pharmacokinetic properties were predicted with Osiris property explorer and PreADMET server. As a result, 18 new ISL analogues (ISL-1 to ISL-18) were synthesized. Among these analogues, ISL-17 showed the strongest inhibitory activities against SGC-7901 and BGC-823 cells, and could induce G2/M cell cycle arrest and apoptosis in these two cell lines. Treatment with ISL-17 resulted in increased ROS production and elevated autophagy levels in SGC-7901 cells. The PI3K/AKT/mTOR signaling pathway was down-regulated after treatment with ISL-17 in SGC-7901 cells. The results of drug-likeness and pharmacokinetic prediction indicated that all the ISL analogues complied with Lipinski's rule of five and Veber rule and had a favorable ADME character. Overall, our results attest that ISL-17 holds promise as a candidate agent against gastric cancer.
