Up-regulation of prohibitin 1 is involved in the proliferation and migration of liver cancer cells

Abstract: Although prohibitin 1 (PHB) is known to be associated with tumors, there are few studies regarding the role of PHB in hepatocellular carcinoma. An earlier glycoproteomics study of ours showed that PHB is over-expressed in MHCC97-H cells, a highly metastatic liver cancer cell line, and can be captured by wheat germ agglutinin. In the present study, western blotting and reverse transcription-PCR experiments showed an approximately 2-fold up-regulation in the expression of PHB in MHCC97-H cells. However, PHB was … Show more

“…We also report the identification of 85 proteins differentially modulated by SGK1, some of which are related to RAN and RANBP1 ( Supplementary Figure S6 ). SGK1 inhibition results in down-regulation of RHOA [ 41 , 42 ], PAK2 [ 43 ]and PHB [ 44 ], all of which are involved in progression through G2/M. These proteins are also over-expressed in conditions associated with liver metastasis, mitotic instability and drug resistance [ 42 44 ].…”

“…However, there is emerging evidence that prohibitin as a protein is required for cell proliferation and adhesion [47] . This protein is also known for activating the Raf-MEK-ERK signaling pathway and inducing cell migration [36] , [48] . Another protein found to be upregulated after treatment with cyclopamine and tamoxifen is keratin 8.…”

“…Higher levels of PHB expression were found in regenerating liver cells, chemically-induced carcinoma, hyperplastic hepatic nodules and hepatocellular carcinomas, and in cancer cells and primary tumor samples (7,2224). The expression of PHB increases in yeast cells during diauxic shift when cells switch from non-oxidative to oxidative metabolism (7).…”
