Unveiling the Link Between Inflammation and Adaptive Immunity in Breast Cancer

Abstract: Inflammation has been recognized as an important driver in the development and growth of malignancies. Inflammatory signaling in cancer emerges from the combinatorial interaction of several deregulated pathways. Pathway deregulation is often driven by changes in the underlying gene regulatory networks. Confronted with such complex scenario, it can be argued that a closer analysis of the structure of such regulatory networks will shed some light on how gene deregulation led to sustained inflammation in cancer. … Show more

“…We decided to analyze and conserve the top-10,000 interactions for the four GCNs in order to have the same size of the four graphs, as well as being able to compare them. Additionally, this network size has been previously observed to be significant to analyze them in terms of structural and functional characteristics (Alcalá-Corona et al, 2016Velazquez-Caldelas et al, 2019;Zamora-Fuentes et al, 2020).…”

“…For this reason, several approaches to reduce its dimensionality have been implemented, including the use of threshold methods, to look for the most significant coexpression relationships (18,21). In particular, in the case of breast cancer molecular subtype networks, the most significant co-expressed pairs have been used as connected nodes in biologically relevant modules (22)(23)(24)(25).…”

“…We observed several low impacted pathways like disease and pathogen-related pathways to be enriched in renal [ 56 , 57 ], and breast [ 58 ] cancer. Breast cancer is the only cancer type that showed enrichment in a number of low-impact cancer-specific pathways and an oncogene pathway (PI3K-Akt signaling [ 59 ]), suggesting this kind of cancer to be especially driven by a high number of prominent oncogenes.…”
