Unusual sequences in the murine immunoglobulin μ–δ heavy-chain region

Abstract: The delta heavy (H) chain of mouse immunoglobulin D (IgD) is unusual both in its structure and in its differential expression relative to immunoglobulin M (IgM; reviewed in ref. 1). The region of DNA between IgM and IgD H-chain constant-region genes is probably implicated in this control. So far only fragments of the area have been sequenced. Now, however, we present the complete sequence as well as the sequence of the introns of the C delta gene. We have found several interesting features (Fig. 1), including … Show more

“…The autoradiographs depicted are from a parallel analysis of WEHI231 and MPCll x WEHI. A restriction map indicates the derivation of the probes for /H-C|X transcripts that avoid repetitive sequences [i and ii (Richards et al 1983)]. Restriction sites: BamHl (B); Bglll (Bg); Hindlll (H); £coRI (R); Sad (S); Xbal (X); Xhol (Xh).…”

“…To examine sequences that might be involved in the differential processing mechanism, the 0.89-kilobase (kb) AccI-KpnI fragment in the middle of the 1.86-kb C4-M1 intron (20) was deleted (pp.AK: Fig. lb).…”

“…Alternating polypurine or polypyrimidine tracts have been previously reported in other eukaryotic genomes such as (CT)25 in sea urchin histone spacer [20] and (CT)3o and (GA)28 in mouse IgC~, [21]. A (CT)9 tract sandwiched with poly(A) and poly(T) observed around a rat calmodulin pseudogene therefore is a new type of simple repetitive sequence.…”
