Osteoporosis, characterized by reduced bone mass and increased bone fragility, is a disease prevalent in women. Likewise, breast cancer is a multifactorial disease and considered the major cause of mortality in premenopausal and postmenopausal women worldwide. Our data demonstrated the association of the MYLK gene and PTGS1 gene variants with osteoporosis and benign breast tumor risk and the impact of ovariectomy on osteoporosis in Korean women. We performed a genome-wide association study (GWAS) of women with osteoporosis and benign breast tumors. There were 60 single nucleotide polymorphisms (SNPs) and 12 SNPs in the MYLK and PTGS1 genes, associated with benign breast tumors and osteoporosis. Our study showed that women with homozygous MYLK rs12163585 major alleles had an increased risk of osteoporosis following ovariectomy compared to those with minor alleles. Women carrying the minor PTGS1 rs1213265 allele and not treated via ovariectomy carried a higher risk of osteoporosis than those who underwent ovariectomy with a homozygous genotype at the major alleles. Our results suggest that both the MYLK and PTGS1 genes are genetic factors associated with the phenotypes, and these associations appear to be modulated by ovariectomy.

Background Breast cancer has become the most commonly diagnosed cancer in the world, but gastrointestinal metastasis from breast cancer is rare. It is extremely rare to have both gastric and sigmoid colonic metastases in one case. For patients considered gastric or colon carcinoma who have a history of breast cancer, it is difficult to identify the origin. Even so, clinicians still need to make accurate diagnosis due to great different treatment principles. Case presentation: We present a case that diagnosed as metachronous gastric and sigmoid colonic metastases from invasive lobular carcinoma of the breast. On June 7, 2021, A 65-year-old female patient came to our department complained recurrent abdominal discomfort for one week. Gastroscopy in our hospital suggested that there was a superficial depressed lesion (type IIC) in her gastric antrum, while pathology indicated poorly differentiated carcinoma. Combined with the patient's breast cancer history and immunohistochemical results, we accurately diagnosed her as gastric metastasis of breast cancer rather than primary gastric cancer. During systematic treatment progress, sigmoid colonic metastasis was detected with colonoscopy on September 19, 2022. Up to May 2023, the patient is still alive with an overall survival greater than 23 months. Conclusions Gastrointestinal metastasis of breast cancer is rare in clinical work, but the incidence of autopsy is up to 15–16%. This phenomenon suggests that some patients with advanced breast cancer have not been accurately diagnosed, which may lead them to receive inappropriate treatment. At present, estrogen/progesterone receptor (ER/PR), GATA binding protein-3 (GATA-3), cytokeratin 7/20 (CK7/20), gross cystic disease fluid protein-15 (GCDFP-15) and mammaglobin are the most diagnostic immunohistochemical markers.

Gastric metastasis from breast cancer is a rare phenomenon in clinical practice, of which the disease features and natural course could be insidious and undistinguished from primary gastric carcinoma. We reported a 37-year-old female patient newly diagnosed as endocrine-dependent breast cancer with synchronous the diffuse thickness of stomach and extensive metastases in the peritoneum as well as bilateral ovaries. The endoscopic view demonstrated a sign of linitis plastic. Histopathological results from gastric biopsy showed molecular features in favor of breast origin, which could be the major source of antidiastole. The prognostic outcomes were heterogeneous among patients with receipt of sequential therapy, where the molecular mechanisms remained to be elucidated.
