The outer-membrane protein ShlB of Serratia marcescens activates and secretes hemolytic ShlA into the culture medium. Without ShlB, inactive ShlA (termed ShlA*) remains in the periplasm. Since Proteus mirabilis L-form cells lack an outer membrane and a periplasm, it was of interest to determine in which compartment recombinant ShlA* and ShlB are localized and whether ShlB activates ShlA*. The cloned shlB and shlA genes were transcribed in P. mirabilis stable L-form cells by the temperature-inducible phage T7 RNA polymerase. Radiolabeling, Western blotting, and complementation with C-terminally truncated ShlA (ShlA255) identified inactive ShlA* in the culture supernatant. ShlB remained cell-bound and did not activate ShlA without integration in an outer membrane. Although hemolytic ShlA added to L-form cells had access to the cytoplasmic membrane, it did not affect L-form cells. Synthesis of the large ShlA protein (165 kDa) in P. mirabilis L-form cells under phage T7 promoter control demonstrates that L-form cells are suitable for the synthesis and secretion of large recombinant proteins. This property and the easy isolation of released proteins make L-form cells suitable for the biotechnological production of proteins.

EinleitungI n der Zeit seit dem Erscheinen des ersten Teiles dieses Sammelreferates (KRAEPELIN 1967) hat der Atmungsdefekt, soweit er sich auf die sogenannte plasmatische ,,petite-Mutation" bezieht, noch keine definitive Kliirung gefunden. Obgleich es zu diesem Thema bereits mehrere zusammenfassende Darstellungen gibt, in denen die zur Zeit wesentlich erscheinenden Fakten diskutiert wurden, mag die in manchem abweichende Betrachtungsweise dieses Referat dennoch rechtfertigen. Insbesondere mag es dazu beitragen, bekannte aber scheinbar unkorrelierte Phiinomene einmal im Zusammenhang zu sehen. In broth cultures thp stabje L-forms of E. coli B and TV 1655 F+ grow as spherical elements with diameters of 0.05 , u up to more than 2 p. Particles below 0.3 p in diameter are probably not, able t o propagate. L-form cells, growing in agar media, are highly pleomorph.Ultrathin sections through L-form cells show that there are no ce!l wall structures. The cells are always surrounded only by the cytoplasmic membrane. Therefore, the L-forms of the two E. coli strains under study may be designa+ed as stable L-forms of the protophst type-UnIike normal rods t h e nucleoids of L-form cells are not marked off clearly from the cytoplasm. There are often vacuoles in the elements, which are surrounded by a typically triple layered membrane. The cytoplasmic membrane may produce local outgrowths of different size. Frequently, intracellular membranes are present in L-form cells. They may be seen as triple layered unit membranes or as st.ructures with 5 and more lagers. With il centre to centre distance of the dark layers of 41 A they are similar t o myelin structures.The quintuple layered structures, which can be found also extracellular, are probably produced by a close contact between the cytoplasmic membrane and the membrane of vacuoles. Formation and function of the intracellular membranes are discussed.Wabxend der Iet,zten Jahre konnten wir mittels Penicillin stabile L-Formen einiger E. coli-Starnme gewinnen (TAUBENECK u.
