Untersuchungen über die Konstitution der Gallensäuren. XXXIX. Mitteilung. Zur Kenntnis der 12-Oxy-cholansäure.

Abstract

Die bisherige Entwicklung der Strukturfrage hat, durch Ausschließung anderer Möglichkeiten, zu der Annahme geführt, daß die beiden im Gesamtgefüge der Molekel noch nicht untergebrachten C-Atome als HC-CH 3 -Gruppe in Eing III. einen Rieber.ring, eingeordnet seien. Um mit dieser Annahme experimentell weiter zu kommen, haben wir uns die Aufgabe gestellt, die Natur dieses Einges, frei von den störenden Einflüssen benachbarter Gruppen, genauer kennenzulernen. Als Vorbild dienten die Methoden, die zum Studium der Einge I und II von Nutzen gewesen waren. Hier hatte die Kenntnis der Oxycholansäuren (3-und 7-) und der entsprechenden Ketosäuren die Möglichkeit geboten, sei es durch oxydative Eingsprengung, sei es auf dem Wege der Bromierung und sich anschließender Eeaktionen, Aufschluß über die Gestaltung dieser Einge sich zu verschaffen. Eing III hat sich bisher nicht in der gleichen Abgrenzung untersuchen lassen, da man noch kein geeignetes, nur in Eing III substituiertes Gallensäurederivat kennt. Denn die fast gleichzeitig von B or s ehe und Todd 1 ) und von Wieland und Deulofeu 2 ) aus Apocholsäure dargestellte /J-Cholensäure 024113302 enthält zwar mit ziemlicher Sicherheit ihre Doppelbindung in Eing III, jedoch in einer Lage, in der sie bisher nicht mit Erfolg zur Konstitutionsermittlung herangezogen werden konnte. Vor allem war sie nicht geeignet, die Gewinnung der zuerst angestrebten Verbindung, der aus der Aufsprengung von Eing III zu erwartenden Tricarbonsäure zu vermitteln.
