Unravelling the core microbiome of biofilms in cooling tower systems

Abstract: In this study, next generation sequencing and catalyzed reporter deposition fluorescence in situ hybridization, combined with confocal microscopy, were used to provide insights into the biodiversity and structure of biofilms collected from four full-scale European cooling systems. Water samples were also analyzed to evaluate the impact of suspended microbes on biofilm formation. A common core microbiome, containing members of the families Sphingomonadaceae, Comamonadaceae and Hyphomicrobiaceae, was found in al… Show more

“…Therefore, interfering with biofilm formation or stimulating its dissociation is an attractive strategy to combat bacterial infections and preventing their chronic development. However, biofilms are not just a major problem in clinics, but also in agriculture because of plant loss due to bacterial diseases ( Ramey et al, 2004 ; Matthysse et al, 2005 ; Koczan et al, 2009 ; Malamud et al, 2012 ; Velmourougane et al, 2017 ), and in industrial water, gas and oil systems, where microbial-induced corrosion causes pipe leakage ( Wang et al, 2013 ; Ramírez et al, 2016 ; Di Gregorio et al, 2017 ).…”

“…Biofilms from full-scale CTs are generally collected from the wall of the CT basins, which is the most accessible location of the system and does not require interruption of operation (Di Gregorio et al 2017). The present pilot study is the first describing analyses of microbial communities from different locations in the recirculation line of a nondisinfected CT.…”

“…No literature is, however, available on the effectiveness of P-limitation in controlling biofouling in cooling water systems. A few studies have investigated the microbiome of biofilms from CTs (Balamurugan et al 2011;Wang et al 2013;Di Gregorio et al 2017;Tsao et al 2019), mainly focusing on MIC and the impact of disinfection, but the effect of nutrient limitations and sample location remains unexplored. In general, further investigation on the effect of nutrient limitations in cooling water would be highly valuable to the industrial field as it guides the selection of feed water, pre-treatment and chemical dosage strategies.…”

“…Biofilm 103 plays a crucial role in Legionella proliferation and survival (Cooper and Hanlon, 2010;104 Flemming et al, 2002;Rogers and Keevil, 1992;Simões et al, 2010). In addition, the 105 composition of the microbial communities in water systems is different in the biofilm and in the 106 water phase (Di Gregorio et al, 2017;Wang et al, 2014). Therefore, analysing the microbial 107 interaction between Lp and the resident microbiota in the water and in the biofilm is warranted to 108 fully understand its life cycle and propose better strategies to control its growth.…”
