Unique Lipid Signatures of Extracellular Vesicles from the Airways of Asthmatics

Abstract: Asthma is a chronic inflammatory disease process involving the conductive airways of the human lung. The dysregulated inflammatory response in this disease process may involve multiple cell-cell interactions mediated by signaling molecules, including lipid mediators. Extracellular vesicles (EVs) are lipid membrane particles that are now recognized as critical mediators of cell-cell communication. Here, we compared the lipid composition and presence of specific lipid mediators in airway EVs purified from the br… Show more

“…According to the hypothesis, enriched KOs indicate that POF patients harboured an impaired inflammation condition. Phosphatidylglycerol, phosphate, L-methionine and pyruvate, whose metabolic enzymes were enriched in the POF group, are known to be related to inflammation and Class II antigen [43,44]. In turn, inflammation promotes the production of acetyl-CoA.…”

“…They also found extensive differences in lipid composition between the EVs of the asthma group and controls. Lipids such as ceramides, ceramide‐phosphates, phosphatidylglycerol and sphingomyelins, which are mediators of inflammation in asthma, were found to be increased in EVs derived from asthma patients …”

“…. A study by Hough et al74 compared the lipid composition and presence of specific lipid mediators in airway EVs purified from the BALF of healthy controls and asthmatic subjects. The results indicated elevated concentrations of EVs in the BALF of asthmatic subjects and that the increase correlated with blood eosinophilia.…”

“…For instance, EVs from non-tumorigenic cells are enriched in prenol lipids, glycerolipids, and fatty acids, while EVs from tumorigenic cells have higher levels of sphingolipids, glycerophospholipids, and sterol lipids [ 76 ]. Similarly, EVs from the bronchoalveolar lavage fluid of asthmatic patients are enriched in ceramide-phosphates, ceramides, and phosphatidylglycerol in comparison to healthy controls [ 77 ]. Even different subtypes of EVs can have distinctive lipidomic profiles.…”
