Unique and Cross-Reactive T Cell Epitope Peptides of the Major Bahia Grass Pollen Allergen, Pas n 1

Abstract: Background: Bahia grass pollen (BaGP) is a major cause of allergic rhinitis. Subcutaneous allergen-specific immunotherapy is effective for grass pollen allergy, but is unsuitable for patients with moderate to severe asthma due to the risk of anaphylaxis. T cell-reactive but IgE nonreactive peptides provide a safer treatment option. This study aimed to identify and characterize dominant CD4+ T cell epitope peptides of the major BaGP allergen, Pas n 1. Methods: Pas n 1-specific T cell lines generated … Show more

“…1). Although dominant allergen epitopes are most simply defined as being those peptides/regions most frequently recognized within the respective allergen sequence 5559, we considered a number of factors in addition to TCL responder frequencies to further refine our selection of epitopes for inclusion in a therapeutic. These included the magnitude of TCL response, number of specific TCL per subject, reproducibility of specific TCL response and ability to target specific T cells in PBMC.…”

“…Assessment of patients receiving temperate GP sublingual immunotherapy (SLIT) tablets showed lowered mean AR medication prescriptions and lower proportions of new onset of asthma than the non‐AIT‐treated groups . However, currently available AIT is not standardised for subtropical GP, which may be important for efficacious treatment of people who are primarily allergic to Panicoideae and/or Chloridoideae GP, given that only one of three T‐cell epitopes is shared between major allergens Pas n 1, Cyn d 1 and Lol p 1 . More specific assessment of GP sensitisation profiles, particularly in countries with multiple climatic zones, may serve to precisely identify the relevant GP to achieve the most efficacious treatment for an individual patient .…”

“…TCC from nonallergic individuals differed, however, from TCC of allergic individuals with respect to their low IL-4/IFN-γ ratio [91,92]. Nevertheless, the value of identifying major T cell epitopes, also from newly identified sources [93], should not be underestimated [94]. It provides a rational basis for the design of modern vaccines for specific immunotherapy which lack potential disease-aggravating, T cell-activating sequences.…”
