Uniplex E6/E7 PCR method detecting E6 or E7 genes in 39 human papillomavirus types

Abstract: We have developed a new human papillomavirus (HPV) assay using the uniplex E6/E7 polymerase chain reaction (PCR) method, which is able to detect E6 and E7 genes in 39 HPV types. We validated the assay for sensitivity and specificity using cloned HPV DNA and clinical samples. A comparative study using Genosearch-31 (GS-31) to determine HPV genotypes in clinical samples was also performed. E6 or E7 genes, measured by uniplex E6/E7 PCR, were detectable in 15 low-risk (HPV-6, -11, -40, -42, -44, -54, -55, -61, -62… Show more

“…The strength of the study was the use of samples in which various HPV genotypes were comprehensively detected with high sensitivity without any cross‐reactivity for each type through uniplex E6/E7 PCR. We confirmed a higher prevalence of multiple infections than reported previously 8 . However, PCR‐based HPV tests are always affected by false‐positives.…”

“…The cells were assayed with Uniplex E6/E7 polymerase chain reaction (PCR), a new HPV‐typing assay method previously established byOkodo et al . The PCR mixture included 1× AmpliTaq Gold ® 360 buffer, 2 mmol/L MgCl 2 , 0.025 U/μL AmpliTaq Gold 360 DNA Polymerase (Applied Biosystems), 2.5 μL DNA, and 0.5 pmol/L primers in a total volume of 25 μL.…”

“…The Uniplex E6/E7 polymerase chain reaction (PCR) assay, established by Okodo et al, (2018) was used for HPV typing. DNA from the residual liquid of cervical cytology specimens (100 μL) was extracted using hot sodium hydroxide and pH adjustment with a Tris solution (HotSHOT) method (Truett et al, 2000).…”
