Unfolding the pleiotropic facades of rosuvastatin in therapeutic intervention of myriads of neurodegenerative disorders

Abstract: The 3-hydroxy-3-methylglutaryl coenzyme A (HMG-CoA) reductase inhibitors, commonly known as statins, are the drugs of choice for the pharmacotherapy of hyperlipidemia as well as for reducing atherosclerotic cardiovascular risk. 1 Statins inhibit the HMG-CoA reductase enzyme and therefore suppress the conversion of HMG-CoA into mevalonate-the first step in cholesterol biosynthesis. Following this step, low-density lipoprotein-cholesterol synthesis is lowered in hepatocytes, which consequently leads to a reduced… Show more

“…Recently, SGLT2is have gained robust traction as a novel class of antidiabetic agents with a unique mechanism of decreasing hyperglycemia by inducing glucosuria [ 12 ]. It does this chiefly by inhibiting the sodium–glucose cotransporter 2 proteins, which resorb glucose in the proximal convoluted tubule of the nephron [ 13 ]. In addition to its primary effect, natriuresis, weight loss, antihypertensive, and antilipidemic effects also occur [ 4 ].…”

“…42 It has pleiotropic effects rather than antihyperlipidemic such as its antioxidant, antiapoptotic effect, and antiinflammatory. 25 Moreover, it induces epigenetic modifications of DNA to protect cells against DNA injury. 26 Because of the previous effects of rosuvastatin, it was chosen to ameliorate CP ovotoxicity, especially it previously ameliorated cisplatininduced nephrotoxicity.…”

“…24 In addition, it exhibits a pleiotropic activity independent of HMG-CoA reductase inhibition, which includes antioxidant, antiapoptotic, and anti-inflammatory effects. 25 Moreover, it induces epigenetic modifications of DNA. These modifications include inhibition of histone and DNA methylation and deacetylation of histone and these changes thought to protect cells against DNA injury and against alteration of tumor suppressor genes.…”

“…Besides, natural statins and antioxidants have multiple pleiotropic effects, such as acting as promoting angiogenesis and neurogenesis, protecting cortical neurons from excitotoxicity, and preventing ischemic stroke. Moreover, they have direct and indirect anti-inflammatory and immunomodulatory effects in inducing pro-inflammatory responses (Banfi et al 2017 ; Bhat et al 2021 ; Husain et al 2019 ). For instance, rosuvastatin administration has been found to significantly reduce NF-κB expression (Husain et al 2017 ).…”
