Unfolded protein response to global ischemia following 48 h of reperfusion in the rat brain: the effect of age and meloxicam

Abstract: The unfolded protein response (UPR) in the hippocampal regions Cornu Ammonis 1 hippocampal region, Cornu Ammonis 3 hippocampal region, and dentate gyrus, as well as in the cerebral cortex of 3-month-old and 18-month-old rats were studied in a model of 15 min of global cerebral ischemia followed by 48 h of reperfusion. UPR was measured by quantifying the protein disulfide isomerase (PDI), C/EBPhomologous protein (CHOP), GRP78 and GRP94 transcripts using qPCR and the amounts of PDI and GRP78 by western blot. The… Show more

“…This difference is consistent with differences observed in the expression of different neurotransmitter system genes (Montori et al 2010a, b, c) or markers of reticulum stress (Llorente et al 2013). Our results show that changes induced by age modify this structuredependent vulnerability.…”

“…Following Ischemia-Reperfusion injury, the PDI expression was enhanced in the cortex even if the statistical significance threshold was not reached (OD ratio: 2.18 ± 0.03). This elevation of PDI expression is reported to be lowered by anti-inflammatory treatment [40]. The hypolipidemic drug treatment before ischemia-reperfusion injury had a lowering effect on the ischemia-reperfusion injury induced PDI surged expression.…”

“…We considered the unfolded protein response (UPR) as a promising target because UPR is activated to restore endoplasmic reticulum (ER) function that is impaired in a variety of stress conditions including stroke, 29 and because the ability to activate UPR declines with age. 1012 …”
