Unexpected Genomic and Phenotypic Diversity of Mycobacterium africanum Lineage 5 Affects Drug Resistance, Protein Secretion, and Immunogenicity

Abstract: Mycobacterium africanum consists of Lineages L5 and L6 of the Mycobacterium tuberculosis complex (MTBC) and causes human tuberculosis in specific regions of Western Africa, but is generally not transmitted in other parts of the world. Since M. africanum is evolutionarily closely placed between the globally dispersed Mycobacterium tuberculosis and animal-adapted MTBC-members, these lineages provide valuable insight into M. tuberculosis evolution. Here, we have collected 15 M. africanum L5 strains isolated in Fr… Show more

“…As previously reported, Rv1979c was deleted independently in all M. africanum lineage 5 strains (del Rv1978-Rv1979c [37]), in our two M. pinnipedii strains (del Rv1964-Rv1979c [38]), and also in more recently derived BCG variants (Rv1964-Rv1988, i.e. RD2) [39,40].…”

“…Similarly, also the closely related human‐adapted M. africanum Lineage six seems to have mostly intact RD5 regions (Fig. ; Brosch et al , ; Brites et al , ; Ates et al , ). Together, these evolutionary observations suggest an advantage for the loss of the ppe38 locus only under specific conditions.…”

“…4. Only one out of 18 M. africanum L5 isolates was found to have an RD5-like deletion (Ates et al, 2018a). In a study by Brodin et al three M. microti isolates from humans had an intact RD5-locus, while four strains isolated from voles had RD5-deletions .…”

“…This convergence largely found in MTB lineages 1, 2, 3 and the MAF lineages, indicates there may be a selective advantage for this. Interestingly, these lineages, compared to MTB L4 strains, have been associated with slower growth and cytokine induction patterns suggestive of immune evasion (8, 14, 65, 8385, 107, 110, 111).…”
