Understanding the Role of Trichoderma reesei Vib1 in Gene Expression during Cellulose Degradation

Abstract: Vib1, a member of the Ndt80/PhoG-like transcription factor family, has been shown to be essential for cellulase production of Trichoderma reesei. Here, we combined transcriptomic and genetic analyses to gain mechanistic insights into the roles of Vib1 during cellulose degradation. Our transcriptome analysis showed that the vib1 deletion caused 586 genes with decreased expression and 431 genes with increased expression on cellulose. The downregulated genes were enriched for Gene Ontology terms associated with c… Show more

“…3f, g and Table 2), which are vital positive TFs that regulate cellulase genes in T. reesei [9,12]. Furthermore, vib1 was prominently enhanced (Table 2), whose deletion caused the decreased expression of almost all cellulase and hemicellulase genes, crucial sugar transporter genes, and main transcriptional activators xyr1 and ace3 in T. reesei [52]. In Neurospora crassa, Vib1 modulated cellulase synthesis by regulating the expression of the essential cellulase regulator CLR2 [53].…”

“…2). Previously, the deletion of these factors was reported to cause loss or reduction of cellulase expression [38][39][40]45 . These factors were also affected by enhanced expression of XYR1 and ACE3 (for example, crt1 is regulated by ACE3 38,47 ).…”

“…In T. reesei , VIB1 is also essential for cellulase production and acts as an important regulator of cellulase induction 36 , 44 . A comparison of the Δ vib1 and Δ xyr1 transcriptomes using cellulose as a carbon source showed a high frequency of overlap between cellulose-induced genes regulated by VIB1 and XYR1 targets 45 . This suggests that VIB1 partially regulates cellulase gene expression via XYR1 45 .…”

“…Fungal transformation was based on infection using the GV3101 strain of Agrobacterium tumefaciens [ 51 ]. T. reesei Rut-C30 (ATCC 56765) [ 52 ] was used as the host strain for genetic transformation. Luria broth (LB) was used to culture E. coli and A. tumefaciens , and MA medium [ 12 ] containing 2% (w/v) glucose and MM [ 46 ] containing 1% (w/v) Avicel or 2% (w/v) glucose were used for general fungal culture.…”
