Understanding the cause of false negative β-d-glucuronidase reactions in culture media containing fermentable carbohydrate

Abstract: Aims:  To explain the basis for false negative β‐glucuronidase reactions seen with culture media containing lactose as a carbon and energy source. Methods and Results:  Escherichia coli strains were assessed for their reactions in culture media containing a β‐d‐glucuronidase substrate either with or without lactose. An assay was developed to test for the expression of β‐d‐glucuronidase at pH 5·0 and pH 7·2. Strains of E. coli that gave false negative glucuronidase reactions on media containing lactose generall… Show more

“…Most bacteria are known to thrive better around neutral pH of 7 (Fricker et al, 2010). As such, the pH ranges obtained in the current study for both tributaries favoured the survival of E. coli as demonstrated by the positive correlation between pH and E. coli counts.…”

“…The Protocol is targeted towards implementation under circumstances without the presence of advanced laboratory infrastructure (e.g., MALDI-TOF for confirmation of species obtained) but nevertheless is expected to give results of high specificity through the use of assays based on the beta-glucuronidase activity of E. coli , which have a high sensitivity and specificity for determining E. coli concentrations in water. “…A wide range of FP and FN rates have been reported (9)(10)(11)(12)(13)(14)(15)48) (Table 3). In TE detection, LTB (or similar methods) generally gave much higher FP rates (Ͼ22%) than enzyme-based media (ϳ10%).…”

“…The rates for FN results are 3.3 to 26.2% and 20.3 to 48.6% for total coliforms and E. coli, respectively (9)(10)(11)(12)(13)(14)(15). The two likely reasons are an inability to recover target organisms from different drinking water matrices and an inability to distinguish target strains from nontarget organisms.…”
