Uncovering the essential genes of the human malaria parasite Plasmodium falciparum by saturation mutagenesis

Abstract: Severe malaria is caused by the apicomplexan parasite Plasmodium falciparum. Despite decades of research the unique biology of these parasites has made it challenging to establish high throughput genetic approaches for identification of therapeutic targets. Using transposon mutagenesis of P. falciparum in an approach that exploited its AT-rich genome we generated >38,000 mutants, saturating the genome and defining fitness costs for 95% of genes. Additionally, this study implied that FIKK10.2 is dispensable, and FIKK10.1 and FIKK9.1 have a low mutagenesis score, suggesting that disruption of these two FIKKs significantly affected parasite fitness (Zhang et al , ). It does remain possible that the global approach is able to detect alterations that partially disrupt but do not delete genes resulting in truncated or disrupted protein expression or function.…”
