Uncoupling of Inflammation and Insulin Resistance by NF-κB in Transgenic Mice through Elevated Energy Expenditure

Abstract: To study the metabolic activity of NF-B, we investigated phenotypes of two different mouse models with elevated NF-B activities. The transcriptional activity of NF-B is enhanced either by overexpression of NF-B p65 (RelA) in aP2-p65 mice or inactivation of NF-B p50 (NF-B1) through gene knock-out. In these models, energy expenditure was elevated in day and night time without a change in locomotion. The mice were resistant to adulthood obesity and diet-induced obesity without reduction in food intake. The adipos… Show more

“…Body composition was measured using quantitative nuclear magnetic resonance (NMR) (Minispec Mn10 NMR scanner, Brucker, Milton, ON, Canada) as described previously (12). In the test, conscious and unrestrained mice were placed individually in a small tube and then placed in the NMR analyzer.…”

“…Finally, abdominal adiposity correlates with plasma C reactive protein levels, indicating that systemic markers of inflammation are also increased with obesity (Brooks et al, 2010). While adipose tissue inflammation in obesity plays a key role in the development of insulin resistance, adipose inflammation in the absence of obesity does not seem to induce insulin resistance (Tang et al, 2010). Thus, it is important to use mouse models with at least some degree of obesity when studying the contribution of individual inflammatory mediators to insulin resistance.…”

“…In this case, IL-6 and PDGF have been reported recently to induce beige cell formation in white fat (66,67). Induction of pro-inflammatory cytokines by over-expression of IKK or NF-kB p65 (RelA) in the fat tissue induces energy expenditure in mice (70,71). Interferon regulatory factor 4 (IFR4) is reported to promote heat production by brown fat through interaction with PGC-1α (68).…”
