“…The application of heteronuclear-based NMR experiments to oligonucleotides has provided many dramatic advantages that had previously been realized with isotopical labeling of protein samples. Uniform isotopic labeling of oligonucleotides now allows the unambiguous assignment of resonances using through-bond correlated NMR experiments, like the HCP Marino et al, 1994a;Varani et al, 1995), HCN (Sklenar et al, 1993(Sklenar et al, , 1994(Sklenar et al, , 1996Farmer et al, 1993Farmer et al, , 1994Tate et al, 1993;Simorre et al, 1995Simorre et al, , 1996Marino et al, 1997), and HCP-CCH-TOCSY (Marino et al, 1994b;Wijmenga et al, 1995), and limits reliance on the more ambiguous NOE-based assignment of sequential connectivity. In addition, the extraction of NOE-derived distance restraints is facilitated by the increased resolution obtained by combining NOESY experiments with evolution periods of heteronuclear chemical shifts Varani et al, 1996;Allain and Varani, 1997).…”

“…For example, sugar proton and carbon resonances can be efficiently assigned by application of HCCH-COSY, -RELAY and -TOCSY experiments Nikonowicz and Pardi, 1993); intraresidue base to sugar connectivities can be made by application of HsCsNbCbHb, HsC~Nb, UbNbCs, HbNbC b and HsC~NbHu experiments (Farmer et al, 1993Sklenfi~ et al, 1993a,b;Tate et al, 1994); HCCH-TOCSY experiments can be used to correlate the H2 and H8 resonances in adenine bases Marino et al, 1994a); and sequential resonance assignments can be made by HCP-type and 13C-31p and ~H-3~P heteroTOCSYtype experiments (Kellogg et al, 1992;Heus et al, 1994;Marino et al, 1994bMarino et al, ,1995Wijmenga et al, 1995). Here we present novel 2D and pseudo-3D triple resonance HNCCCH experiments that provide through-bond NH/ NH2 to H6 connectivities for uridine and cytidine base protons in r3C-/~SN-labeled RNAs.…”

“…6A shows the Cl'-Hi' cross peaks in a 2D constant time heteronuclear single quantum correlation (HSQC) (20,21). The proton chemical shifts are correlated to the glycosidic nitrogens in a HCN (22,23) experiment (Fig. 6B), which uses the two consecutive transfers of magnetization from 1H to 13C to '5N and then back.…”
