Unaltered [3H]GBR-12935 binding after chronic treatment with dopamine active drugs

Abstract: Rats were injected intraperitoneally with haloperidol 0.5 mg/kg, raclopride 1 mg/kg, bromocriptine 2.5 mg/kg, d-amphetamine 2.5 mg/kg, or cocaine 10 mg/kg twice daily for 21 days. The animals were sacrificed 72 h after last injection. Control rats were injected with saline, following the same schedule. The radioligand [3H]GBR-12935 was used as a presynaptic marker for dopamine neurites. There were no significant differences in [3H]GBR-12935 binding to striatum between drug-treated rats and controls.

“…Consistent with the findings of the current study, most studies have shown that chronic, intermittent cocaine treatment also failed to alter ligand binding to the dopamine transporter (Allard et al 1990;Kula and Baldessarini 1991). A decrease in dopamine transporter density has been associated with chronic cocaine, but only after 10 days of withdrawal (Sharpe et al 1991).…”

“…The diagnostic cathegory of depression did not influence the results, neither did antidepressant medication nor the method of suicide. Concerning the individuals treated also with neuroleptics, several studies have revealed unchanged binding parameters related to dopamine uptake sites after treatment with these drugs (Allard et al, 1990b;Valchar et al, 1989). It has been shown that striatal dopaminergic D2 receptors are subject to alterations in depression.…”

“…Since chronic neuroleptic administration does not affect DAT density (Allard et al 1990;Ase et al 1999;Reader et al 1998;Rivest et al 1995), there was no rationale to withhold treatment prior to the [ ] ␤ -CIT study, eight of the schizophrenic patients were treated with antipsychotic medication at the time of scanning. The other subjects were off-medications for an average of 17.5 Ϯ 11 days (range 7-33 days).…”
