Umbelliferone Improves an Impaired Glucose and Lipid Metabolism in High-Fat Diet/Streptozotocin-Induced Type 2 Diabetic Rats

Abstract: Umbelliferone (UMB) is a natural product that has several pharmacological effects including antihyperglycemic activity in diabetic rats. Thus, the objective of this study was to investigate the effect of UMB on insulin resistance and on the regulation of glucose and lipid metabolism in type 2 diabetic rats. Type 2 diabetes was induced in rats by feeding a high-fat diet (45 kcal% fat) and a single dose of streptozotocin injection. After 8 weeks of treatment, UMB significantly reduced the elevated blood glucose … Show more

“…The results of liver pathological section by haematoxylin and eosin staining showed reduced fat accumulation in the liver. Furthermore, expression of peroxisome proliferator‐activated receptor γ (PPARγ) and surface GLUT4 in adipose tissue was upregulated, suggesting that umbelliferone could enhance insulin sensitivity and promote GLUT4 translocation through activation of PPARγ. In type 1 diabetes, oxidative stress is an important factor which worsens pancreatic β‐cell dysfunction .…”

“…In Africa and Asia, S. alata leaves and flowers are currently used to regulate lipid absorption, obesity, and fat levels in blood serum. Aqueous leaf extract considerably reduced blood glucose levels, serum cholesterol, triglyceride, hepatic triglyceride, serum leptin, and insulin levels in Wistar mice [57].…”

“…Peroxisome proliferator-activated receptor γ (PPAR γ ) is an essential nuclear hormone receptor acting on the transcription of genes (e.g., glucose transporter gene GLUT4) involved in glucose disposal. A study finding reveals that umbelliferone upregulated the expression of surface GLUT4 gene and PPAR γ in adipose tissue, enhancing insulin sensitivity leading to increased peripheral glucose uptake [ 55 ]. Osthole also activated PPAR γ , increasing insulin sensitivity, and facilitated the synthesis and storage of fat [ 56 ].…”
