Ultraviolet Sensitivity Gene of Escherichia coli B

Abstract: The ultraviolet sensitivity gene of Escherichia coli B was introduced into a K-12 recipient by transduction with phage P1. The uvs gene of E. coli B is cotransducible with the proC locus of K-12, is closely linked to tsx, is not linked to lacZ, and only rarely to purE. The transductants are mucoid, filamentous on irradiation, and show plating-medium response. The order of markers is lacZ proC tsx uvs purE.

“…This mutant was Plkc sensitive and also T7 sensitive, The mutation to mucoid (excess capsule formation) can occur at two distinct loci: capR (synonomous with Ion; see references 3,14,19) and capS (21). The capR mutants are also UV sensitive (3,9,14). ES370 was not UV sensitive and assumed to be a capS mutation.…”

“…The receptors for phages T3, T4, and T7 are in the lipopolysaccharide layer of the cell wall, and the T2 and T6 receptors are in the lipoprotein (30,31). In E. coli B, which cannot become mucoid (9), no cross-resistance is found among the T2, T6, and the T3, T4, T7 groups. It has been shown in E. coli B that resistance to phages T3, T4, and T7 (B/3, 4, 7) is due to a change in the lipopolysaccharide (32).…”

“…Although the specificity ratios for B1-8 IgM and separately expressed i and X are very similar, lower concentrations of NIP and NP are required for inhibition of separately expressed X and X compared to B1-8 IgM. (23,24), so that the absence of this protease either acting upon p, or being responsible for activating other proteases that act on p, may be the major factor resulting in accumulation in E. coli B.…”

“…Although these treatments may rescue a variety of strains [5] they were first demonstrated, and exert their largest effect, on E. coli strain B . This strain carries a mutation, lon [6], conferring a reduced ability to re-initiate cell division following U.V. irradiation, resulting in the formation of filaments and death.…”
