Ultraviolet inactivation of murine leukemia virus and its assay in permissive and non‐permissive cells

Abstract: This study confirms the different titration patterns of murine leukemia viruses (MuLV) ( 2 ) Infection by MuLV of its restrictive host was enhanced by the UV-inactivated homologous MuL V. ( 3 ) UV-inactivated NB-tropic virus propagated in N-type cells enhanced plaque formation of N-tropic virus in B-type cells and not that of B-tropic virus in N-type cells. UV-inactivated NB-tropic virus propagated in B-type cells enhanced plaque formation of B-tropic virus in N-type cells, but not that of N-tropic virus in … Show more

“…The gene does not restrict the chemical activation of endogenous leukemia virus of the nonpermissive host, but does restrict perpetuation of the activated virus (5,14,15). In cell culture, the resistance is not absolute and high virus concentrations can overcome the restriction (3,4,7).…”

“…Resistance is dominant and F1 hybrids restrict both types of virus. The Fv-1 gene is also expressed in cultured cells but the restriction is not absolute, and titration of the restrictecl virus yields two-hit kinetics (4)(5)(6)(7). While cells of F1 hybrids restrict both N-and B-tropic viruses, a class of viruses designated NB-tropic are not restricted by either allele.…”

“…Earlier experiments on Fv-1 restriction involved titration patterns o n cells from a variety of mouse strains (Yoshikura, 1973;Decleve ef al., 1975;Jolicceur and Baltimore, 1975;Pincus eb a/., 1975) including NIH, the original prototype Fv-1" mouse cell. Experiments were therefore performed to characterize Fv-1 restriction in NIH/3T3 cells and in another Fv-I" cell line, C3H 10T1/2.…”

“…Lactic dehydrogenase virus in the FLV stock was disregarded because it does not disturb genetic studies. B-tropic LLV, WN1802-B (Rowe and Hartley, 1972), was propagated either on cultured Y H-7 (Yoshikura, 1973), an established cell line of C57B1/6 origin, or o n BALB/c embryo cells. The titers of the LLV from the two sources were near.…”
