Ultraviolet A and B affect human melanocytes and keratinocytes differently. A study of oxidative alterations and apoptosis

Abstract: Ultraviolet (UV) radiation is an etiologic agent for malignant melanoma and non-melanoma skin cancer, but the spectral range responsible for tumor induction is still to be elucidated. In this study, we compared effects of UVA and UVB irradiation on normal human melanocytes (MCs) and keratinocytes (KCs) in vitro. We demonstrate that UVA irradiation induces immediate loss of reduced glutathione (GSH) in both MCs and KCs. Exposure to UVA also causes reduced plasma membrane stability, in both cell types, as estima… Show more

“…Piceatannol has previously been reported to increase the GSH level in B16 melanoma cells, 27) while UVB irradiation have no effect on the GSH level in melanocytes. 28) Our finding that GSH levels increased in keratinocytes is important for UV protection in skin.…”

“…Primary keratinocytes were obtained from foreskin circumcisions of fair-skinned donors (0-2 years of age), and pure cultures of keratinocytes were established and cultured as previously described (Andersson et al, 2001;Larsson et al, 2005).…”

“…As extensive UV radiation causes melanocytes to undergo apoptosis (35,36), we further studied the effects of Ref-1 on cell viability with UVB radiation. After seeding into plates overnight, primary normal melanocytes were infected by Adhgal and Adref-1, respectively, for 16 hours (200 MOI).…”
