Ultrastructure, protein synthesis and secretion of day-6 rabbit blastocysts cultured in a chemically defined, protein-free medium

Abstract: Day-6 rabbit blastocysts were cultured in Ham's F10 medium supplemented with polyvinylpyrrolidone as a macromolecular component, for 4 to 12 h. The integrity of the blastocyst cells was demonstrated by electron microscopy. Expansion and biosynthesis of proteins and of DNA were studied after culturing in the presence of 35S-methionine and 3H-thymidine. Polyvinylpyrrolidone did not interfere with the subsequent protein analysis, which was performed by two dimensional gel electrophoresis followed by silver staini… Show more

“…6) reflecting a possible glycoprotein. A similar set of proteins also has been described by Henkel et al (1990), although they were detectable in a more acidic area (4.6-5.4). This may be explained by the use of ampholines by Henkel et al (1990) instead of immobilines.…”

This may be explained by the use of ampholines by Henkel et al (1990) instead of immobilines. This set of glycoproteins can be explained as transforming growth factor 'e', a large fragment of acrogranin (Parnell et al 1992;Diazo-Cueto et al 2000), which has been shown to run at 25 kDa (Parnell et al 1990(Parnell et al , 1992.…”

“…The suitability of this medium for the culture of rabbit blastocysts has previously been demonstrated (Henkel et al 1990). Addition of bFGF to the culture medium induced primitive streak formation in day 6 blastocysts within 24 h. This represents a slight acceleration in comparison to development in vivo, and could possibly be caused either by an overstimulation or by a premature stimulation of embryonic disc development in the culture system.…”

“…On the other hand, the seven mentioned placental proteins of low molecular weight are clearly absent in the pre-implantation embryo (Henkel et al 1990;Hrab6 de Angelis and Kirchner 1993). Their nature has not yet been identified.…”
