Oogenesis Histology UltrastructureThe present study was carried out to describe the ovary and oogenesis in the freshwater bivalve Caelatura parreyssi from histological and ultrastructural points of view. Ten adult healthy clams collected from the River Nile were used in this study. The specimens were dissected and gonads were obtained and processed for histological and ultrastructural examination. Histologically, the ovary of C. parreyssi is composed of numerous of follicles which are connected together by connective tissue. Each follicle contains different stages of developing oocytes; oogonia, previtellogenic oocytes, vitellogenic oocytes and postvitellogenic oocytes (mature oocytes). At ultrastructural level, stages of female gametogenesis were examined; the process of oocytes formation and yolk deposition were described in successive stages of oogenesis. The structure and function of auxiliary cells and follicle cells in ovarian follicles during vitellogenesis were discussed. In conclusion, this study showed that the follicle cells play an important role during oogenesis but probably are not the major source of yolk precursors. The vesicular connective tissue is probably the main source of nutrients for vitellogenesis. The auxiliary cells appear to play an integrate role in the development of the oocytes as their functions can permit a transfer of precursors necessary to vitelogenesis.

The level of vitellogenin‐like (Vg‐like) proteins in the hemolymph of the soft‐shell clam (Mya arenaria) was indirectly determined using an alkali‐labile phosphate (ALP) measurement assay. Biochemical characterization of hemolymph from actively reproducing clams showed the presence of Zn, Ca and of substantial amounts of ALP, all of which are indicative of fish serum vitellogenin, reported to be a Zn and Ca containing glycolipophosphoprotein. ALP assays undertaken on different phosphate‐containing compounds (phosphate‐buffer saline, diphospholipids and 12,000×g supernatants of Vg‐rich trout ovary homogenates) showed little interference by the first two substrates and confirmed the specificity of the ALP assay as an indirect method of determining Vg‐like proteins in clam hemolymph. Moreover, clams injected with 17β‐estradiol and two known xenoestrogens (nonylphenol and pentachlorophenol) significantly induced hemolymph ALP levels. Field work revealed significant differences in hemolymph ALP levels from clams collected at specific intertidal stations of the Saguenay Fjord during temporal (May to October) and spatial (June) surveys in 1997, which appear to indicate that (anti‐)estrogenic chemicals may be present in the Saguenay Fjord. Our preliminary results thus show that this simple and rapid ALP assay holds promise as a biomarker of estrogen disruption with molluscan shellfish as bioindicators. ©1999 John Wiley & Sons, Inc. The aim of the present article was to investigate the oogenic cycle of Mytilus galloprovincialis sampled in the Bay of Naples, and to immunolocalize 3β‐hydroxysteroid dehydrogenase (3β‐HSD), 17β‐hydroxysteroid dehydrogenase (17β‐HSD), and P450 aromatase, enzymes involved in the synthesis of two sex hormones: testosterone and 17β‐estradiol. We demonstrate that the oogenic cycle starts in late summer‐early fall and continues in early winter when the first event of spawning occurs; other spawning events take place until June, when the ovary is spent and contains a few empty ovarian follicles and numerous somatic cells, that is, adipogranular cells and vesicular connective tissue cells. During the oogenic cycle, apoptotic events occur at the level of oogonia, previtellogenic oocytes, as well as follicle cells; by contrast, necrosis events probably take place in vitellogenic oocytes, which, once degenerated, transfer their content to healthy oocytes. Finally, the present data demonstrate that 3β‐HSD, 17β‐HSD, and P450 aromatase are present in the ovary both during the reproductive and nonreproductive phases.
