Ultrastructural Visualization of Surface Carbohydrate Structures on Mycoplasma Membranes by Concanavalin A

Abstract: Surface carbohydrates of Mycoplasma mycoides var. capri were made visible by the cytochemical staining procedure with concanavalin A, horseradish peroxidase, and diaminobenzidine.

“…As a model system to perform sensitive measurements of protein binding with different concentrations of protein, we used the specific binding of the glycoprotein concanavalin A (Con A) to ribonuclease B (RNase B) immobilized on a monolayer (11‐mercaptoundecanoic acid), as shown in Figure a. Con A, an important member of the legume lectin family, is widely used in biology and biochemistry to characterize glycoproteins and other sugar‐containing entities on the surfaces of various cells . The specific binding of Con A and RNase B has become a standard affinity model for assessing biosensor performance.…”

“…1A). Therefore, most experiments were performed with mycoplasmas prefixed with glutaraldehyde before harvesting and cytochemical labeling in order to stabilize the membranes' gross structure and to prevent redistribution and cluster formation of the ferritin granules (14,29).…”

“…In pursuing our previous studies (27)(28)(29) on the surface architecture of mycoplasma membranes, we investigated with the electron microscope the localization, distribution, and chemical nature of the anionic sites on the surfaces of intact mycoplasmas and isolated mycoplasma membranes. As a visual probe, we used a polycationized derivative of ferritin (2) which binds electrostatically to anionic sites on the surface of either glutaraldehyde-fixed or unfixed membranes.…”
