Purpose To ascertain possible cell damage from cryopreservation, the ultrastructure of human oocytes cryopreserved by slow cooling was assessed. Materials and methods Cryopreservation was performed through two protocols with one-step or two-step propanediol. Fresh control oocytes were examined for comparison. Samples were processed for transmission electron microscopy analysis. Results By light microscopy, both fresh and frozen-thawed oocytes appeared regularly rounded, with intact zona pellucida, and homogeneous cytoplasm. By electron microscopy observation, organelles were abundant and uniformly dispersed. Mitochondria-smooth endoplasmic reticulum associations appeared regular. However, both the amount and density of cortical granules appeared abnormally reduced in frozen-thawed samples. Slight to moderate vacuolization was also found in the ooplasm of oocytes of both frozen groups. Conclusions Slow cooling ensures a good overall preservation of human oocytes. However, cytoplasmic vacuolization and cortical granule loss appears associated with cryopreservation, irrespective of the protocol used.

Para investigar os efeitos da ingestão de diferentes quantidades da planta Phalaris angusta em bovinos, oito bezerros, com idade variando entre 6-8 meses, foram divididos em 4 grupos com 2 animais cada. Os animais do grupo I receberam somente P. angusta na alimentação, enquanto que os animais do grupo II receberam P. angusta (75%), aveia (Avena sativa) e azevém (Lolium multiflorum) (25%). Os animais do grupo III receberam P. angusta (50%), aveia e azevém (50%) e os animais do grupo IV receberam somente aveia e azevém e serviram como controles. Todos os animais que ingeriram P. angusta adoeceram. Um animal do grupo I morreu 34 dias após o início da ingestão da planta e os outros animais foram sacrificados, in extremis, em um período que variou de 18 a 32 dias após o início do experimento. Os principais sinais clínicos observados foram alterações de locomoção, tremores generalizados, quedas e crises convulsivas. Alterações macroscópicas foram observadas apenas no encéfalo e eram caracterizadas por focos de coloração verde-azulada no tálamo, mesencéfalo e medula oblonga. Microscopicamente observou-se pigmento granular marrom-amarelado no citoplasma de neurônios das regiões macroscopicamente afetadas. Alterações ultra-estruturais consistiram de lisossomos contendo material com densidade e orientação variáveis. A quantidade de P. angusta ingerida não foi um fator determinante na gravidade do quadro clínico, nem na intensidade das lesões observadas. A intensidade dos sinais clínicos também não teve uma relação direta com a severidade das lesões macro e microscópicas. Phalaris angusta demonstrou ter ação exclusivamente neurotóxica e deve ser considerada no diagnóstico diferencial em casos de animais com sinais clínicos de origem nervosa, consistentes com síndrome tremorgênica.

Abstract. Lead (Pb) poisoning of a pregnant heifer was diagnosed based upon clinical signs (head pressing, blindness, muscle twitching) and a blood lead concentration of 1.73 ppm. Blood and urinary Pb half-lives with CaNa 2 EDTA/thiamine therapy were determined to be 2.08 and 1.38 days, respectively. Many cations (Ca, Fe, Zn, Na, Cu), including Pb, were excreted at higher concentrations in urine during therapy. Blood (0.425 ppm) and liver (4.85 ppm) Pb concentrations in the fetus were 71.7% and 84.3% of the same tissue Pb concentrations of the dam, indicating a significant transfer of Pb in utero. Severe polioencephalomalacia was described in the adult, and hepatic lysosomes with metallic electron densities were present in the fetus.A severely depressed and dehydrated approximately 10-month-old angus heifer (175 kg, estimated) was presented to the Mississippi State University, College of Veterinary Medicine (CVM) Animal Health Center. The herd had recently experienced 2 acute yearling deaths due to unknown causes. This heifer had been found standing in a pond and was reluctant to move. Clinical signs included head pressing, staggering, muscle twitching, and lack of a menace response. Acute toxicosis (lead [Pb], organic pesticides, mercury) or a vitamin deficiency (vitamin A, thiamine) were suspected. Other rule outs included rabies, polioencephalomalacia, thromboembolic meningoencephalitis, listeriosis, and other less common central nervous system (CNS) differential diagnoses. 21 Blood was taken for a complete blood count (CBC) with differential, serum chemistry panel, and blood Pb concentration analysis. Initial therapy (day 0) consisted of thiamine hydrochloride a (1 g, intramuscular [im]), dexamethasone b (20 mg, intravenous [iv]), and oral electrolyte solution c (10 liters via stomach tube).The following morning the neurological signs were more pronounced and included, in addition to those previously noted, hyperesthesia, ptyalism, and bruxism. Blood Pb concentration was 1.73 ppm (CVM Diagnostic Services, Toxicology Laboratory, normal < 0.35 ppm) and the CBC and clinical chemistries were unremarkable. Day 1 treatments were 10 liters electrolyte solution per os, CaNa 2 EDTA d (13 g in 250 ml of saline iv), thiamine hydrochloride (1 g, im), and acepromazine maleate e (18 mg, im
