Ultrastructural and molecular analyses of homozygous-viable Drosophila melanogaster muscle mutants indicate there is a complex pattern of myosin heavy-chain isoform distribution.

Abstract: We describe the ultrastructural and initial molecular characterization of four homozygous-viable, dominantflightless mutants of Drosophila melanogaster. Genetic mapping indicates that these mutations are inseparable from the known muscle myosin heavy-chain (MHC) allele Mhc^, and each mutation results in a muscle-specific reduction in MHC protein accumulation. The indirect flight muscles (IFMs) of each of these homozygous mutants fail to accumulate MHC, lack thick filaments, and do not display normal cylindrica… Show more

“…This result is intriguing in that we previously documented another difference between the two TDT cell types in Mhc 7 and Mhc 9 mutants (O'Donnell et al, 1989). The mutants display normal numbers of thick filaments in the TDT muscle large cells but a reduced number in the small cells.…”

“…Myosin Expression Levels-Transgenic lines were crossed into the Mhc 10 background, and expression levels were determined by SDS-polyacrylamide gel electrophoresis as described previously (29). Six dissected upper thoraces from 2-day-old female transgenic flies were homogenized in 60 l of SDS gel buffer.…”

“…There are generally 26-28 large cells and four small cells (O'Donnell et al, 1989;Peckham et al, 1990). The cells are visualized in paraffin sections cut horizontal to the axis of the muscle (Fig.…”
