Ultrastable gold substrates for electron cryomicroscopy

Abstract: Despite recent advances, the structures of many proteins cannot be determined by electron cryomicroscopy because the individual proteins move during irradiation. This blurs the images so they cannot be aligned with each other to calculate a 3D density. Much of this movement stems from instabilities in the carbon substrates used to support frozen samples in the microscope. Here we demonstrate a new gold specimen support that nearly eliminates substrate motion during irradiation. This increases the sub-nanometer… Show more

“…Au/Au grids were prepared as described in the literature 33 . Briefly, grids were prepared by first coating C-flat (Protochips, Inc., Morrisville, NC) CF-1.2/1.3-2Au 200 mesh holey carbon grids with ~50 nm of gold using an Edwards Auto 306 evaporator.…”

“…To address these issues, the samples were vitrified using a semi-automated Spotiton V1.0 robot 15,16,36 , a novel device for preparing cryoEM samples that uses piezo-electric dispensing to apply small (50pL) drops of sample across a “self-blotting” nanowire grid as it flies past en route to plunge into liquid ethane. The nanowire grids, manufactured in house, used either lacey carbon or lacey gold supports 37 ; the gold support helps to optimize image quality when acquiring data with the grid tilted relative to the electron beam 14,38 . Tilted data collection was necessary to address highly preferred particle orientation observed during initial data collections.…”

“…For subsequent experiments (RyR-30 μM Ca 2+ , RyR-300 μM Ca 2+ -10 μM Ryanodine), holey-gold grids were prepared as described by Russo and Passmore (Russo and Passmore, 2014) from either C-flat CF-1.2/1.3-2C-T (Protochips Inc, NC) or from Quantifoil R1.2/1.3 (Quantifoil Micro Tools GmbH, Germany). 3 μL of each sample was applied to the holeygold grids after plasma cleaning with a mixture of H 2 and O 2 .…”
