Purpose: We investigated validation and optimization of ultrasound-assisted dispersive liquidliquid microextraction (UADLLME) as a preparation method for detection of methadone in saliva samples. Methods: We used blank and methadone-containing saliva samples and also standard methadone solution. Sodium hydroxide and chloroform were added to samples and they were held in ultrasonic bath. Then preparations were centrifuged and extracted analyte was analyzed by gas chromatography-mass spectrometry (GC-MS). Accuracy was measured by Intra and between-day mean relative errors (RE). Precision was assessed by coefficient of variation (CV). Recovery, specificity, linearity and limits of detection and quantification were also determined. Optimization was conducted for ultrasound duration, pH and extraction phase volume. Efficiency of dispersive liquid-liquid microextraction (DLLME) and UADLLME were compared. Results: Intra and between-day accuracies (2.3 -7.5%), recovery (89.4-115.5%) and precision (5.2-11.3%) were all acceptable. Calibration curve was linear in the concentration range of 150 ng/mL-10 µL/mL with R2 >0.9995 and equation of y=86.901x-5342.5. Limits of detection and quantification were 50 and 150 ng/mL, respectively. Specificity was measured by comparing retention times of saliva samples (containing methadone metabolites and other commonly used drugs) during UADLLME/GC-MS analysis and no interference was observed. Recovery of UADLLME was 1.4 of DLLME. Solvent and sample volumes required for UADLLME were 1/200 and 1/20 of DLLME. The greatest efficiency obtained at pH of 10, with ultrasound treatment duration of 5 minutes and extraction phase volume of 1000 µL. Conclusion: Study found that UADLLME/GC-MS is a valid and efficient method for detection of methadone in oral fluid.

A novel and rapid ultrasound- and salt-assisted liquid-liquid extraction coupled with high-performance liquid chromatography has been optimized by response surface methodology for the determination of oleuropein from olive leaves. Box-Behnken design was used for optimizing the main parameters including ultrasound time (A), pH (B), salt concentration (C), and volume of miscible organic solvent (D). In this technique, a mixture of plant sample and extraction solvent was subjected to ultrasound waves. After ultrasound-assisted extraction, phase separation was performed by the addition of salt to the liquid phase. The optimal conditions for the highest extraction yield of oleuropein were ultrasound time, 30 min; volume of organic solvent, 2.5 mL; salt concentration, 25% w/v; and sample pH, 4. Experimental data were fitted with a quadratic model. Analysis of variance results show that BC interaction, A(2) , B(2) , C(2) , and D(2) are significant model terms. Unlike the conventional extraction methods for plant extracts, no evaporation and reconstitution operations were needed in the proposed technique.

BACKGROUND: A novel and rapid method for the separation and simultaneous extraction of two types of polysaccharides from kelp was developed using an ultrasonic-assisted solid-liquid extraction method. Many polysaccharides obtained from diverse plant sources have important properties, such as anticancer, antioxidant and antitumor activities. Edible brown algae contain many bioactive compounds, including polysaccharides, vitamins and minerals. As the most common brown alga on the coastal areas of the Korean peninsula, kelp was selected to extract laminarin and fucoidan. Using ethanol as a solvent, the effects of ultrasonic-assisted solid-liquid extraction on the extraction of polysaccharides from kelp, such as the extraction period, temperature, pH and mass:solvent ratio were investigated by high performance liquid chromatography.RESULTS: Compared to traditional heating and microwave-assisted extraction methods, ultrasonic-assisted solid-liquid extraction produced higher extraction yields in a shorter time. The optimal conditions for the ultrasonic-assisted solid-liquid extraction supporting the highest recoveries of the two polysaccharides were 40°C, ethanol as a solvent at pH 6, 40 min and a 1:30 g mL −1 mass:solvent ratio. Under the optimal conditions, the recoveries of fucoidan and laminarin were 94.29% and 88.90%, respectively. The real quantities of fucoidan and laminarin extracted from kelp with synthesized carboxymethylcellulose sodium Schiff bases added to ethanol in the proposed method were 120.7 ± 0.26 and 45.3 ± 0.19 mg g −1 , respectively. CONCLUSION: Ultrasonic-assisted solid-liquid extraction not only improved the recoveries of the extraction, but also was an eco-friendly, efficient and promising alternative method for the simultaneous extraction of polysaccharides from kelp.
