Ultrasonic Self-Emulsification Nanocarriers for Cellular Enhanced Astaxanthin Delivery

Abstract: Oil in water (O/W) nanocarriers were prepared for cellular enhanced astaxanthin delivery using a (3-carboxypropyl) triphenylphosphonium bromide (TPP)-modified casein by an ultrasonic self-emulsification method. The nanocarriers of casein emulsion loaded with astaxanthin and casein modified by TPP emulsion encapsulated with astaxanthin were 227 and 543 nm, respectively, with a spherical shape. The thermal stability and resistance to ultraviolet (UV) radiation ability of astaxanthin were significantly improved a… Show more

“…This was mainly because ultrasound could change the LG structure and introduce more hydroxyl groups, and the increase of hydroxyl groups could significantly improve the anti-oxidation ability. In conclusion, ultrasound could improve the antioxidant activity of LG and this was similar to the conclusion reached by Zhang et al when studying the changes of antioxidant activity when protein binds to astaxanthin [25] .…”

“…MMP Analysis. MMP is an important indicator of cell health and is often associated with intracellular oxidative stress, 41 The change of intracellular MMP is usually used to evaluate the antioxidant capacity of delivery systems. JC-1 forms aggregates with red fluorescence in healthy cellular mitochondria with a high membrane potential.…”

“…The operation steps were as follows: human corneal epithelial cells (HCEC) were seeded in 96-well plates, then incubated with a fresh medium containing different concentrations of lutein and lutein/NC-8 nanoparticles for 24 h. Subsequently, MTT was added and incubated for 4 h to check the cell viability. 5…”

“…3,4 In addition to the above advantages, an ideal carrier should also have good biocompatibility, controlled release, targeted delivery, and synergistic function with functional factors. 5,6 Among them, size is also an essential factor affecting the bio-logical performance of carriers. For example, small particle size will increase the targeting performance and residence time of nanocarriers.…”
