Ultra-trace element analysis of human follicular fluid by ICP-MS/MS: pre-analytical challenges, contamination control, and matrix effects

Abstract: Human follicular fluid was collected from 56 women undergoing IVF treatment and analyzed for 11 (ultra-)trace elements using a “dilute and shoot” ICP-MS/MS method.

“…Between 0.5 mL and 5.0 mL FF was aspirated from a single large follicle in each ovary. The follicular fluid was transferred to a sterile Petri dish, and after the oocyte removal, the fluid was placed into a 15 mL conical tube and centrifuged for 5 minutes at 1500 g, distributed into 1.8 mL polypropylene cryo-vials, and stored at 80 • C until further use (11) .…”

“…One FF aliquot from each sample (n = 197) was analysed for As, Hg, Cd and Pb, at the Laboratory of Inorganic and Nuclear Chemistry, Wadsworth Center, New York State Department of Health (Albany, NY, USA). The analytical method, described in detail in a previous publication ( Galusha et al ., 2019 ), was developed on an Agilent 8900 inductively coupled plasma tandem mass spectrometer (ICP-MS/MS) (Agilent Technologies, Santa Clara, CA, USA). Briefly, specimens were prepared along with standards and quality control samples (QCs), diluted into polypropylene autosampler tubes and analysed by ICP-MS/MS.…”

“…It is worth emphasising that besides the mentioned biofluids and tissues, virtually any biospecimen can be interrogated in ICP-MS regarding elemental profiles and a disrupted homeostasis. In the recent years, a range of studies started exploring rather untraditional specimens including faeces [ 80 ], tears [ 81 ], milk [ 82 ], breath [ 83 ], skin [ 84 ], bone marrow fluid [ 85 ], nasal exudate [ 86 ], follicular fluid [ 87 ] or seminal plasma [ 88 ] to just name a few examples. Especially microsampling of biological fluids seems to become more relevant to ensure minimal invasive procedures and to target scarce fluids.…”
