Ultra-tough injectable cytocompatible hydrogel for 3D cell culture and cartilage repair

Abstract: A high-performance hydrogel was synthesized by a facile dual dynamic crosslinking strategy that showed injectability, cytocompatibility, broadly tunable mechanical properties and the potential for repair of load-bearing tissues.

“…A,B) Plots of the compressive versus tensile strength and modulus for BC–PVA–PAMPS (this work) and other strong hydrogels (see Table S1 in the Supporting Information for data and references). [ 13–35 ] The multiple data points for BC–PVA–PAMPS are for different compositions. C) BC–PVA–PAMPS easily bears the weight of a 100 lb.…”

“…As depicted in Figure 1, two kinds of water-soluble naturally derived polymers, carboxymethyl chitosan (CMC) and oxidized alginate (OSA), were chosen to fabricate injectable hydrogels. The oxidized alginate (OSA) with aldehyde groups was obtained via the periodate oxidation reaction, modified on our previous work [30]. The oxidation reaction was confirmed by 1 H-NMR spectrum of SA and OSA ( Figure S1).…”

“…The dynamic gathering of CPBA could lead to a self-reinforcing effect inside the hydrogel matrix, resulting in high compressive and tensile moduli of the hydrogel over 1.0 MPa, including the highest compressive modulus up to 5.6 MPa. Moreover, after 3 months of implantation of this hydrogel into osteochondral defect in a rabbit model, smooth and complete cartilage layer was obtained, and the enrichment of glycosaminoglycan (GAG) and type-II collagen were observed in the cartilage layer (Zhao et al, 2018).…”

“…Hydrogels crosslinked by ionic bonds interactions could also be sufficiently robust for cartilage regeneration, which was examined on rabbit model bearing articular osteochondral defect. Through the generation of ionic interaction and borate bonding, poly(vinyl alcohol) (PVA) was crosslinked by 4carboxyphenylboronic acid (CPBA) to bridge the polymer chains with the presence of calcium ions (Zhao et al, 2018). The dynamic gathering of CPBA could lead to a self-reinforcing effect inside the hydrogel matrix, resulting in high compressive and tensile moduli of the hydrogel over 1.0 MPa, including the highest compressive modulus up to 5.6 MPa.…”
