Ultra‐performance Liquid Chromatography–High‐definition Mass Spectrometry Analysis of Constituents in the Root of Radix Stemonae and those Absorbed in Blood after Oral Administration of the Extract of the Crude Drug

Abstract: It is concluded that the screening capability of MetaboLynx(TM), together with the utilisation of MS(E) in structural elucidation, can facilitate a rapid and comprehensive detection and effective structural characterisation of Stemona alkaloids in Radix Stemonae extract, which could be applied to other Chinese medicine. The results provide helpful chemical information of Radix Stemonae extract for further pharmacology and active mechanism research.

“…Fortunately, the multivariate data mining method (MMA) has proved to be helpful in elucidating the effective constituents in herbal medicines in order to assess and control the overall quality. 12 The experimental setup for LC/MS coupled with the multiple data processing approach is shown in Fig. 8 LC-MS has enjoyed a growing popularity as a platform for herbal medicine studies because its high throughput, so ionization, and good coverage of metabolites offers a robust, reliable and economical method for quantitative constituent analysis.…”

“…All of these herbs were confirmed as being able to regulate the neuroendocrine hypofunction89101112131415, which is closely related to multiple disordered metabolic pathways of “Kidney-Yang deficiency syndrome” (KYDS) such as damage to the hypothalamic-pituitary-target gland (adrenal, thyroid, and gonad) axis1617181920. However, the therapeutic mechanism, effective constituents, and potential targets of AS1350 effects on KYDS are unclear.…”

“…Compounds may be functional by oxidation, hydrolysis or reduction; primary metabolites may also undergo conjugation reactions to form secondary metabolites. 34, 35 Our results show that the BSNXD-derived serum increases ER β expression in HUVECs, and results in an enhanced NO and reduced MDA production. ER β other than ER α antagonist can block these effects induced by the drug-derived serum; meanwhile, NOS inhibitor can significantly decrease the NO production in HUVECs.…”
