Ultra-low-dose estrogen therapy for female hypogonadism

Abstract: In females, endogenous estrogen secretion increases gradually before pubertal development. The benefits of low-dose estrogen therapy in patients with Turner syndrome were originally discussed by Ross et al. and Quigley et al. These seminal studies used ethinyl estradiol (EE2), starting at a dose of 25 ng/kg/d. We hypothesized that the initial dosage of estrogen could be titrated to more closely mimic physiological increments of endogenous estrogen. Therefore, our recent study initiated EE2 treatment at a dosag… Show more

“…Many research findings have supported the idea that EE2 might have certain negative consequences, including headaches, breast tenderness, nausea, dizziness, and weight gain. Therefore, EE2 is often advised for oral administration at a lower doses to avoid its side effects [48,49].…”

“…At the beginning of menopause, the production of estrogen drops significantly and thus leads to well-known symptoms in women in this phase of life [166]. Bone health, heart health, female fertility, various regulatory effects on glucose metabolism, the maintenance of immune function, nerve function and the role of estrogens in the treatment of many pathologies are also modulated by estrogen signaling [114,[127][128][129][166][167][168][169][170][171][172][173][174][175][176][177][178][179][180][181]. The estrogen circulating in the woman's body acts in an endocrine, paracrine or intracrine manner and binds to the three most important of its receptors, the estrogen receptor α (ERα), the estrogen receptor β (ERβ) or the G protein-coupled estrogen receptor 1 (GPER1) [182,183].…”
