Ultra-fast searching assists in evaluating sub-ppm mass accuracy enhancement in U-HPLC/Orbitrap MS data

Abstract: A strategy, detailed methodology description and software are given with which the mass accuracy of U-HPLC-Orbitrap data (resolving power 50,000 FWHM) can be enhanced by an order of magnitude to sub-ppm levels. After mass accuracy enhancement all 211 reference masses have mass errors within 0.5 ppm; only 14 of these are outside the 0.2 ppm error margin. Further demonstration of mass accuracy enhancement is shown on a pre-concentrated urine sample in which evidence for 89 (342 ions) potential hydroxylated and g… Show more

“…Noteworthy, the average error was approximately zero around m/z 622.0289, the m/z of the compound used for lock-mass calibration. The m/z deviations depend on m/z value (Mihaleva et al, 2008;Lommen et al, 2011). We followed the plausible argument of the authors to assume that we must have correctly annotated a certain number of lipid sum formulae because they are known to exist in our sample.…”

“…Both q-TOF and Orbitrap mass analyzers generally have mass accuracies below 5 ppm [54][55][56][57][58] and have been routinely employed in the analysis of food. While the specified measured mass accuracies for these instruments are sufficient for non-targeted analysis, impaired data quality can occur, especially when analyzing complex sample matrices [58].…”

“…Initial capabilities are limited to targeted search capabilities in preprocessed metabolomics data. Metabolomics platforms for which analysis tools are provided are LCMS [28,29], GCMS and GCGCMS. Preprocessed data can be acquired using the MetAlign software and stored in MADMAX.…”
