Uji Sitotoksik Ekstrak Etanol Kulit Batang Awar-awar dengan Metode BSLT

Abstract. Cancer is a disease that is a major health problem in the world. Pulai leaves (Alstonia scholaris (L.) R. Br.) are part of a plant that is thought to have anticancer potential because it’s contain secondary metabolites such as alkaloid. Alkaloid compounds are known to have activity that can inhibit growth and kill cancer cells. This study aims to determine the cytotoxic activity and LC50 value of the ethanol extract, water fraction, ethyl acetate fraction, and n-hexane fraction of pulai leaves. The cytotoxic test was carried out using the Brine Shrimp Lethality Test (BSLT) method on Artemia franciscana Kellogg shrimp larvae and probit analysis to determine the LC50 value. The test concentrations used were 10 ppm, 100 ppm, 250 ppm, 500 ppm, 750 ppm, and 1000 ppm. Testing the cytotoxic activity of the ethanol extract, water fraction, ethyl acetate fraction, and n-hexane fraction of pulai leaves produced LC50 values of 207,2526 ppm, 282,1630 ppm, 187,5858 ppm, and 484,2839 ppm, respectively. This implied that the ethanol extract, water fraction, ethyl acetate fraction, and n-hexane fraction of pulai leaves have a cytotoxic effect and have anticancer potential as indicated by an LC50 value of less than 1000 ppm. Abstrak. Kanker merupakan salah satu penyakit yang menjadi masalah kesehatan utama di dunia. Daun pulai (Alstonia scholaris (L.) R. Br.) merupakan bagian tanaman yang diduga memiliki potensi sebagai antikanker karena mengandung golongan senyawa metabolit sekunder seperti alkaloid. Golongan senyawa alkaloid diketahui memiliki aktivitas yang dapat menghambat pertumbuhan dan membunuh sel kanker. Penelitian ini bertujuan untuk mengetahui aktivitas sitotoksik dan nilai LC50 dari ekstrak etanol, fraksi air, fraksi etil asetat, dan fraksi n-heksan daun pulai. Uji sitotoksik dilakukan dengan metode Brine Shrimp Lethality Test (BSLT) terhadap larva udang Artemia franciscana Kellogg dan analisis probit untuk mengetahui nilai LC50. Konsentrasi uji yang digunakan adalah 10 ppm, 100 ppm, 250 ppm, 500 ppm, 750 ppm, dan 1000 ppm. Pengujian aktivitas sitotoksik ekstrak etanol, fraksi air, fraksi etil asetat, dan fraksi n-heksan daun pulai menghasilkan nilai LC50 berturut-turut sebesar 207,2526 ppm, 282,1630 ppm, 187,5858 ppm, dan 484,2839 ppm. Hal ini menunjukkan bahwa ekstrak etanol, fraksi air, fraksi etil asetat, dan fraksi n-heksan daun pulai memiliki efek sitotoksik dan berpotensi sebagai antikanker yang ditandai dengan nilai LC50 kurang dari 1000 ppm.
