Ubiquitylome Analysis Reveals a Central Role for the Ubiquitin-Proteasome System in Plant Innate Immunity

Abstract: Protein ubiquitylation profoundly expands proteome functionality and diversifies cellular signaling processes, with recent studies providing ample evidence for its importance to plant immunity. To gain a proteome-wide appreciation of ubiquitylome dynamics during immune recognition, we employed a two-step affinity enrichment protocol based on a 6His-tagged ubiquitin (Ub) variant coupled with high sensitivity mass spectrometry to identify Arabidopsis proteins rapidly ubiquitylated upon plant perception of the mi… Show more

“…These include serine-glyoxylate aminotransferase (SGAT/ AGT1), alanine-glyoxylate aminotransferase 2 (AGT2), glycolate oxidases (GOX1 and GOX2), glutamate-glyoxylate aminotransferases (GGT1 and GGT2), as well as PMDH2 that provides NADH to the photorespiratory enzyme hydroxy pyruvate reductase 1 (HPR1). Most of these proteins were detected in a ubiquitomics study of 2-week-old Arabidopsis seedlings treated with flg22, a microbe-associated molecular pattern (MAMP) peptide (Ma et al, 2021), suggesting a role for ubiquitination in adjusting peroxisomal photorespiratory activities during immune response.…”
