U-box E3 ubiquitin ligase PUB8 attenuates abscisic acid responses during early seedling growth

Abstract: ABSCISIC ACID-INSENSITIVE3 (ABI3) and ABI5 are two crucial transcription factors in abscisic acid (ABA) signaling, and their homeostasis at the protein level plays a decisive role in seed germination and subsequent seedling growth. Here, we found that PLANT U-BOX 8 (PUB8), a U-box E3 ubiquitin ligase, physically interacts with ABI3 and ABI5 and negatively regulates ABA responses during early Arabidopsis (Arabidopsis thaliana) seedling growth. Loss-of-function pub8 mutants were hypersensitive to ABA-inhibited c… Show more

“…Moreover, PUB44 down‐regulates the osmotic stress response by triggering the degradation of CPK4 (Fan et al, 2023). During seedling growth, PUB8 negatively regulates two transcription factors, ABI5 and ABI3, attenuating the ABA response (Li et al, 2023). In this regard, it may be possible that CaPUB24 mediates the degradation of various types of proteins, including cytoplasmic ABA receptors, transcription factors, and protein kinases, involved in ABA signalling.…”

“…Total RNA was extracted by using Trizol reagent (Invitrogen). The RT-qPCR analysis was conducted as described (He et al, 2023; Li et al, 2023). In brief, 1.0 μg DNase-treated RNA was reverse transcribed in a reaction volume of 20 μl containing oligo-(dT)19 primer and Moloney murine leukemia virus reverse transcriptase (Thermo Fisher Scientific).…”

“…The plasmids were transformed into Agrobacterium tumefaciens strain GV3101 and injected into tobacco ( N. benthamiana ) leaves. As described previously (He et al, 2023; Li et al, 2023), YFP and 4,6-diamidino-2-phenylindole (DAPI) fluorescence of infected leaves were observed under a confocal laser scanning microscope (Olympus) after 48 h of co-expression. Primers used for cloning are listed in Supplemental Data Set 1.…”

“…Furthermore, ubiquitination-mediated protein degradation is an important process in ABA signal transduction. [ 20 , 21 ]. In regulating the expression of stress-responsive genes, transcription factors, post-transcriptional modification and secondary metabolism play a vital role in responding to abiotic stress [ 22 , 23 ].…”
