Tyrosine Phosphorylation of c-Maf Enhances the Expression of IL-4 Gene

Abstract: Maf proteins are involved in a variety of biological processes, such as oncogenesis, lens development, and differentiation. In immune system, c-Maf transactivates IL-4 promoter, and ectopic expression of c-Maf skews primary T cell response toward the Th2 pathway. Numerous transcription factors are subjected to posttranslational modification. In this study, to our knowledge, we show for the first time that c-Maf is subjective to tyrosine phosphorylation in Th cells and that the level of its tyrosine phosphoryla… Show more

“…27 In MM cells, both c-Maf and MafB can be phosphorylated by the Ser/Thr kinase GSK3, thus increasing MM cell proliferation and colony formation. 28 The ubiquitination of c-Maf could lead to its degradation which could possibly decrease its oncogenicity in MM cells.…”

“…Furthermore, SATB1 coordinates Th2 cell differentiation by positively regulating Gata3 (Notani et al, 2010). SATB1, NFIL3, MAF, and PPARG positively regulate the gene expression of the Th2 cytokine Il4 (Ribeiro de Almeida et al, 2009;Kashiwada et al, 2011;Lai et al, 2012;da Rocha Junior et al, 2013;Kwon et al, 2014). In addition to the positive effects on Th2 cell differentiation, NFIL3 suppresses Th17 cell development by directly binding and repressing the Rorg promoter .…”

“…To evaluate the effects of PD-L1 expression by liver cancer cells on cytotoxic T-cell function, CD8+ T cells were purified from spleens and lymph nodes of 6-to 8-week-old male BALB/c mice using the EasySep Mouse CD4 and CD8a Positive Selection Kit (StemCell Technologies, Vancouver, BC, Canada) and enriched as previously described [25]. The enriched CD4+ and CD8+ T cells were first rested in complete RPMI 1640 medium for 24 h, then co-cultured with parental and PD-L1-expressing BNL-MEA cells in different ratios for cytokine and cell proliferation assays.…”
