Tyrosine kinase signaling in and on the endoplasmic reticulum

Abstract: Tyrosine kinases are signaling molecules that are common to all metazoans and are involved in the regulation of many cellular processes such as proliferation and survival. While most attention has been devoted to tyrosine kinases signaling at the plasma membrane and the cytosol, very little attention has been dedicated to signaling at endomembranes. In this review, I will discuss recent evidence that we obtained on signaling of tyrosine kinases at the surface of the endoplasmic reticulum (ER), as well as in th… Show more

“…These included ER and endosomal signals, which may suggest RTKs to localize in some extent to various membrane compartments. Endosomal and ER localization may be due to both physiological activity (identified for many RTKs; for reviews, see Miaczynska, 2013 ; Fraser et al , 2017 ; and Farhan, 2020 ) and some unspecific antibody staining.…”

“…PKDCC regulates the transport of ERP29, RP44, BiP, and PDIA3 by phosphorylating transport receptors. These proteins are components of ER proteostasis and play an important role in UPR-induced apoptosis ( 44 ). Therefore, FAM20C, FAM198A, and PKDCC are essential in regulating ER proteostasis and UPR signaling.…”
