“…Among the different stages, discs at 3 w were found to have the most abundant and active ECM expression. These discs showed high expression levels of both the early-stage markers Col3a1 and Col5a1 and the late-stage markers Comp and Dcn, which are vital for the structural stability of the ECM and mechanical strength sensing of fibrocartilage [35][36][37][38] . In other words, discs at this stage, while growing in size via cell proliferation, also become sturdier in order to respond to mechanical force by secreting specific ECM components.…”

“…Importantly, the increases in Col2𝛼1 and Acan, as the main ECM proteins in matured cartilage, and decreases in MMP13 and Col1a2, as important indicators for the differentiation of mature chondrocytes toward hypertrophy, illustrated that chondrocytes better maintained their phenotype on the soft micropillar substrate. In addition, the increase of Col5𝛼l, that plays a vital role in mediating the progenitor cell niche of cartilage, [45] and Col6𝛼1, that locates at the direct vicinity of chondrocytes and protects cartilage from degradation via proteolysis resistance, [46] further elucidated the phenotype maintenance on soft micropillar substrates, although the basic expression levels of these two proteins cannot be compared to Col2𝛼1 at all. [47,48] We then performed qPCR and confirmed these gene changes of Col2a1, Acan, MMP13, and Col1a2 (Figure 2i).…”

“…Collagens are the most common family, with more than 20 distinct collagen types recognized currently [ 97 , 98 , 99 , 100 ]. Collagens are essential for the production of extracellular matrix fibrillar and microfibrillar networks, basement membranes, and other extracellular matrix structures [ 101 , 102 ].…”
