Type-Specific Cell Line Models for Type-Specific Ovarian Cancer Research

Abstract: BackgroundOvarian carcinomas consist of at least five distinct diseases: high-grade serous, low-grade serous, clear cell, endometrioid, and mucinous. Biomarker and molecular characterization may represent a more biologically relevant basis for grouping and treating this family of tumors, rather than site of origin. Molecular characteristics have become the new standard for clinical pathology, however development of tailored type-specific therapies is hampered by a failure of basic research to recognize that mo… Show more

“…As expected, in SKOV3 cells which express wild-type B-Raf, GA did not elicit a reduction in the level of B-Raf protein (Fig. 1A) (34). The level of Raf-1, also an Hsp90 client, declined only after 10 h of GA treatment (Fig.…”

“…A similar GA-mediated reduction in MLK3 protein levels was observed in HEY1B and TOV21G ovarian cancer cell lines, which express wild-type B-Raf (see Fig. S2A in the supplemental material) (34). Taken together, these results indicate that the GA-dependent decline in MLK3 protein levels is closely correlated with an increase in Hsp70 protein levels and a deactivation of ERK and JNK signaling.…”

“…8;accessed 15 June, 2014). NRAS mutations were found in 6% of malignant ascites/washings from patients with ovarian cancer (43); in 1 case (2.5%) of HGSC (44); and one case of LGSC from which a cell line has also been derived (45). In the paired samples tested, both the invasive and adjacent borderline compartments contained the same NRAS mutation.…”

“…LGSC cell lines and murine models are limited (35,36), and the RM cells were used as a model for LGSC on the basis of their dependence on K-Ras mutation. Nevertheless, the RM cells form aggressive disease, in contrast to the slower disease progression of LGSC in women.…”
