Type IV Secretion Machinery Promotes Ton-Independent Intracellular Survival of Neisseria gonorrhoeae within Cervical Epithelial Cells

Abstract: Survival of Neisseria gonorrhoeae within host epithelial cells is expected to be important in the pathogenesis of gonococcal disease. We previously demonstrated that strain FA1090 derives iron from a host cell in a process that requires the Ton complex and a putative TonB-dependent transporter, TdfF. FA1090, however, lacks the gonococcal genetic island (GGI) that is present in the majority of strains. The GGI in strain MS11 has been partially characterized, and it encodes a type IV secretion system (T4SS) invo… Show more

“…Some bacteria have more than one tonB or tonB-like gene, and in all cases, the connection between TonB-ExbBD and iron acquisition, when examined, has been clear (1,4,10,29,39,58,59,64,93,97,121,126,139). But recently, it has become apparent that some bacteria that contain an OM and IM, including L. pneumophila, and species of Chlamydia, Chlamydophila, Coxiella, Ehrlichia, Francisella, and Rickettsia do not have TonB-ExbBD (29,74).…”

“…Named VirB6 in A. tumefaciens and the related systems, these homologues are also inner-membrane proteins and interact with other proteins of their T4SSs (55)(56)(57). A. tumefaciens VirB6 interacts directly with inner-membrane protein VirB8 and stabilizes VirB3 and VirB5 in the secretion apparatus (63,64,66,67). A. tumefaciens VirB6 can be cross-linked to DNA using the TrIP method (58).…”

“…Intracellular growth in the absence of inorganic iron was assayed using a modification of the method described by Zola et al (42). ME180 cervical epithelial cells were seeded at a density of 2.5 ϫ 10 5 cells/ml to a 12 well plate and grown to 70 to 80% confluence at 37°C with 5% CO 2 in McCoy's 5A medium (Gibco) supplemented with 10% heat-inactivated fetal bovine serum (HI-FBS) (Gibco).…”

“…The seventh strain we identified as possessing a distinct GGI type during the course of this study. The GGI-encoded T4SS in N. gonorrhoeae has been shown to secrete DNA active in natural transformation and to facilitate TonB-independent iron uptake during intracellular growth of gonococci (42). Thus, the presence of the T4SS genes in Mc might affect horizontal gene transfer or intracellular growth of N. meningitidis strains possessing the T4SS genes.…”
