Type IV Collagen, Type IV Collagenase Activity and Ability of Cell Proliferation in Human Thyroid Tumours

Abstract: These findings suggest that staining for type IV collagen and type IV collagenase activity reflect the ability of cell proliferation, and help predict the aggressiveness of invasion and metastasis in human thyroid tumours.

“…We believe that the following hypothesis explains why HTTs show this characteristic von Kossa staining pattern. It has been already demonstrated by immunostaining that collagen type IV is abundantly present in the stroma of HTT . The stroma of HTTs was positive for collagen type IV immunostaining in this study.…”

“…Kossa positivity seemed to be observed especially around small vessels in HTT stroma. Immunostaining for type IV collagen reveals that there is a basement membranous constituent in HTT stroma, because type IV collagen is a component of the basement membrane and HTT stroma has been reported to be composed of type IV collagen . Kossa‐positive abundant tiny black dots were distributed in the basement membranous component.…”

“…The activity of base membrane COL4 indicates the cell proliferation and metastatic abilities of thyroid cancer (46). Kusunoki et al (47) showed that the level of COL4 collagenase in patients with thyroid cancer was higher, compared with that in the normal thyroid, and COL4 may assist in predicting the invasion and metastasis of thyroid cancer. The expression of COL5A1 is suppressed by microRNA-145 in anaplastic meningionas (48).…”

“…Multiple enzymes are involved in basement membrane degradation including the matrix metalloproteinases (MMPs), a group of nine Zn +2 ‐dependent endopeptidases. The main component of basement membrane architecture is type 4 collagen, making MMP‐2 and MMP‐9 the main foci in the study of basement membrane degradation 18 . MMPs are balanced by the tissue inhibitors of MMPs, which act to prevent MMP protein digestion.…”
