Type I Interferon Acts as a Major Barrier to the Establishment of Persistent Infectious Bursal Disease Virus Infections

Abstract: Infectious bursal disease virus (IBDV), the best characterized member of the Birnaviridae family, is a highly relevant avian pathogen causing both acute and persistent infections in different avian hosts. Here, we describe the establishment of clonal, long-term, productive persistent IBDV infections in DF-1 chicken embryonic fibroblasts. Although virus yields in persistently-infected cells are exceedingly lower than those detected in acutely infected cells, the replication fitness of viruses isolated from pers… Show more

“…The standard curve included viral genome concentrations ranging from 5 Â 10 7 to 5 copies, considering that 1 PFU corresponds to one viral genome, from which an equation with an R 2 of $0.99 was generated for the calculation of the absolute number of mRNA copies (per 12.5 ng total yield of RNA subjected to reverse transcription reaction) by linear regression, which was normalized to the relative amount of rRNA 28S. This procedure was adapted from that described earlier (69). All the primer sequences (Table 3) were specific for PRV strain Kaplan (GenBank accession number JQ809328) and were obtained from Tombácz et al (70).…”

“…Most viral infections trigger a series of signaling cascades leading to the expression of type I IFNs, which exerts key roles in cellular antiviral responses (29,30). The cGAS-STING DNA sensing pathway has been reported to play a pivotal role in inducing the production of IFNs and suppressing the replication of viruses, in particular DNA viruses (31).…”

“…Also, higher GFP expression was observed in TRIM25 KO DF-1 cells infected with NDV-GFP and VSV-GFP recombinant viruses. Previous results from our laboratory showed that IFN plays a key role in apoptotic cell death of IBDV-infected cells ( Cubas-Gaona et al, 2018 ; Broto et al, 2021 ). Significantly, reduced apoptosis triggering was observed after IBDV infection of TRIM25 KO DF-1 cells when compared with the parental DF-1 cells.…”
